ABSTRACT
AbstractEMMPRIN (extracellular matrix metalloproteinase inducer, CD147) participates in the progression of various malignancies by stimulating synthesis specific metalloproteinases (MMP) from peritumoral fibroblasts. In present study, expression and functional role EMMPRIN was investigated pancreatic neoplasm. QRT-PCR, immunohistochemistry, immunoblot, ELISA analyses were used to analyze expression, localization, release EMMRPIN. Silencing performed using siRNA oligonucleotides, consequences assessed growth assays, invasion as well MMP1/MMP2 VEGF ELISA. mRNA levels 2.2-fold increased cancer (n=52) 2.0-3.5-fold other neoplasm (n=105), but unchanged chronic pancreatitis (n=10) compared normal tissues (n=9). Strong predominantly membranous immunostaining observed cells surrounding stromal cells. serum also significantly patients (n=44) (4.13 +/- 0.28 ng/ml) with an AUC 0.97 healthy volunteers (n=29) (0.95 0.16 ng/ml; p
