SFRP1CpG island methylation locus is associated with renal cell cancer susceptibility and disease recurrence

ABSTRACT
Loss of the secreted Fzd-related protein 1 (SFRP1) and concurrent alteration SFRP1/WNT pathway are frequently observed in human cancers such as renal cell cancer (RCC). Whether methylation a SFRP1 CpG island locus normal solid tissues is associated with increased tissue specific risk has not been determined to date. Here we measure attributable DNA tissue. Pyrosequencing bisulfite treated was used for case-control study including 120 normal-appearing autopsy specimens 72 obtained from tumor adjacent areas, cross sectional 96 RCCs. Association demographic factor age, clinicopathological parameters course patients investigated. We show significant hypermethylation RCC compared kidney tissues. Inter quartile analysis revealed 6-, 13- 11-fold second, third fourth quartiles age matched subgroup (p = 0.001, p 1.3E-6, 6.9E-6). Methylation tumors an independent predictor recurrence free survival. methylation, accumulates risk, suggesting this CGI sub region epigenetic susceptibility RCC. Our data underline need further analyze risks conferred by methylated loci development cancers.
