ABSTRACT
BACKGROUND/OBJECTIVES: Colitis is a serious health problem, and chronic obesity associated with the progression of colitis.The aim this study was to determine effects natural raw meal (NRM) on high-fat diet (HFD, 45%) dextran sulfate sodium (DSS, 2% w/v)-induced colitis in C57BL/6J mice.MATERIALS/METHODS: Body weight, colon length, weight-to-length ratio, were measured directly.Serum levels obesity-related biomarkers, triglyceride (TG), total cholesterol (TC), low density lipoprotein (LDL), high (HDL), insulin, leptin, adiponectin determined using commercial kits.Serum pro-inflammatory cytokines including tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-6 detected ELISA kit.Histological performed hematoxylin eosin (H&E) staining assay.Colonic mRNA expressions TNF-α, IL-1β, IL-6, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) by RT-PCR assay.RESULTS: weight biomarkers (TG, TC, LDL, HDL, adiponectin) regulated prevented NRM treated mice.NRM significantly suppressed shortening reduced ratio HFD+DSS induced mice (P < 0.05).Histological observations suggested that edema, mucosal damage, loss crypts HFD DSS.In addition, decreased serum cytokines, inhibited these iNOS COX-2 mucosa 0.05). CONCLUSION:The results suggest has an anti-inflammatory effect against DSS-induced mice, are due amelioration and/or inflammatory reactions.
