ABSTRACT
Myeloid differentiation-2 (MD-2) has been associated with endotoxin and inflammatory disorders because it can recognize lipopolysaccharide (LPS) binding attenuate Toll-like receptor 4 (TLR4)-mediated signaling. However, its role in allergic inflammation yet to be clarified. We examined whether single nucleotide polymorphisms (SNPs) MD-2 promoter affect expression aimed clarify the relationship between Der p 2 allergy SNPs of promoter.The function effects cytokines immunoglobulin on secretion mRNA were investigated 73 subjects different gene variants. Peripheral blood mononuclear cells cultured or without LPS presence Dermatophagoides pteronyssinus group allergen (Der 2), followed by extraction cytokine analysis. The culture supernatants collected for measurement.Patients (rs1809441/rs1809442) had increased expressions MD-2, ε heavy chain IgE (Cε), interleukin (IL)-8; however, only IL-8 further up-regulated after stimulation. Patients tended have high levels IL-1β, IL-6, IL-8, IL-10, tumor necrosis factor (TNF)-α Increased secretions IL-10 found stimulation, an IFN-γ decreased IL-4 noted stimulation.The proinflammatory secreted predetermined (rs1809441/rs1809442). Through LPS, these may serve as indicator pathological phenotype 2-induced inflammation.
