ABSTRACT
Neuroblastoma, an embryonic tumor of the sympathetic nervous system, is deadliest type cancer, accounting for 6-9% all childhood cancers. The omega (ω)-6 [linoleic acid (LA)]: ω-3 [eicosapentaenoic (EPA)]=13/1 ratio associated with presence chronic disease, and it has been reported that reducing this to 7/1 protects against cancer cardiovascular diseases. This study aimed investigate anticancer effects different ratios ω-6 fatty acids in human neuroblastoma cells (SHSY-5Y). SHSY-5Y were treated 48 72 h. viability acid-treated was measured using methylthiazolyldiphenyl-tetrazolium bromide. percentage cell apoptosis detected Fluorescein Isothiocyanate-conjugated Annexin-V/PI assay, reactive oxygen species (ROS) analysis performed flow cytometry. expression levels necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), transforming growth factor beta 1 (TGFβ1) genes determined real-time polymerase chain reaction. EPA LA separately significantly inhibited proliferation SH-SY5Y within h (p<0.001). It found decreased groups due application polyunsaturated at ratios, most effective dose ω-3:ω-6 ratio: 1/1. ROS compared control group lowest level observed 1/4 group. Both TNF-α, IL-6, TGFβ1 mRNA expressions increased after addition mixtures control; they decrease increasing ratio. first examine on cells. ω ratios. In contrast, when ω-3: increased, amount ROS. Additionally, as increases, a release pro-inflammatory cytokines noted.
