Archaeal Life on Tangkuban Perahu-Sampling and Culture Growth in Indonesian Laboratories

ABSTRACT
The aim of the expedition to Tangkuban Perahu, West Java was obtain archaeal samples from solfatara fields located in Domas crater. This one places, where scientists University Regensburg Germany had formerly isolated Indonesian archaea, especially Thermoplasma and Sulfolobus species but not fully characterized. We collected five mud holes with temperatures 57 88 °C pH 1.5-2. A portion each sample grown at modified Allen's medium 80 °C. From four out enrichment cultures were obtained, autotrophically on elemental sulphur heterotrophically sulfur yeast extract; electron micrographs are presented. In laboratories Universitas Indonesia isolates cultured 55-60 order grow tetraetherlipid synthesizing both Thermoplasmatales Sulfolobales. Here, we succeeded culture same type cells, which been Regensburg, probably a Freundt's medium, species. harvested cells documented by phase contrast microscope equipped digital camera. Our next steps will be further characterize genetically Perahu isolates.
