ABSTRACT
To assess the new GenoType® MTBDRplus assay for rapid detection of multidrug-resistant tuberculosis (MDR-TB) in comparison with DNA sequencing to identify drug resistance mutation profiles China.Using MTBDRplus, susceptibility testing (DST) and sequencing, 237 Mycobacterium strains were tested.The sensitivity was 75.0% (126/168) isoniazid (INH) resistant strains, 93.5% (157/168) rifampicin (RMP) strains. It correlated well 94.9% 99.6% agreement each strain category 100% specificity all categories. The two most common rpoB mutations S531L (53.6%, 90/168) D516G (17.3%, 29/168) RMP-resistant INH dominated by katG 315 locus (S T, N, R, I) (73.7%, 124/168), a rare mutation, S315N (6.5%, 11/168), not covered identified. combination inhA-15/inhA-8 katG315 (34 strains) characteristically displayed MDR-TB (23.5%), but INH-monoresistant strains.Although Genotype is reliable molecular test detecting MDR-TB, significant proportion China contain that would be missed assay. Further improvements may achieved incorporating this into increase China.
