Structural changes in the kidneys after long–term use of NSAIDs and new cyanothioacetamide derivatives

ABSTRACT
Purpose of the study. The aim study was to evaluate differences in morphometric parameters kidneys rats with longterm administration nonsteroidal anti-inflammatory drugs and new derivatives cyanothioacetamide assess nephrotoxicity. Materials methods. synthesized at “ChemEx” Research Laboratory were preliminarily subjected virtual bioscreening. We selected 5 heterocyclic compounds potential analgesic activity, which confirmed various pharmacological tests. To chronic effect on kidneys, administered intragastrically a dose mg/kg for 14 days. comparison nimesulide (5 mg/kg), indomethacin (7 acetylsalicylic acid paracetamol 50 mg/kg. stereometric indices assessed each group. Results. revealed that laboratory codes d02–123, d02–133, d02–139, d02–172, d02–149 have lower nephrotoxicity compared reference drugs. In particular, compound code showed minimal changes structure proximal distal tubules absence hemorrhages, indicating its low risk kidneys. Derivatives d02–123 d02–133 adaptive changes, such as an increase area decrease volume normal glomeruli, may be associated compensatory mechanisms during long–term exposure. Conclusions.
