ABSTRACT
This study was aimed to investigate the effects of DNA methylation inhibitor 5-aza-2'-deoxycytidine (5-Aza-CdR) and histone deacetylase trichostatin A (TSA) on DLC-1 gene transcription regulation molecular biological behaviours in human multiple myeloma RPMI-8226 cells. The cells were treated respectively with 5-Aza-CdR TSA alone, or both combination; cell proliferation apoptosis, expression, protein expression Ras homolog family member (RhoA) Ras-related C3 botulinum toxin substrate 1 (Rac1) examined by CCK-8 method, RT-PCR ELISA, respectively. results showed that had growth inhibitory apoptosis-inducing dose-dependent manner (P < 0.05). Compared a single drug (5-Aza-CdR alone), significantly enhanced after treatment combination weakly expressed control group; alone its re-expression dose-dependently plus significantly.Compared control, decreased RhoA Rac1 It is concluded can effectively reverse cells; has synergistic effect re-expression. have significant These may be related inhibition Rho/Rho kinase signalling pathway.
