Deciphering molecular heterogeneity and dynamics of human hippocampal neural stem cells at different ages and injury states

ABSTRACT
While accumulated publications support the existence of neurogenesis in adult human hippocampus, homeostasis and developmental potentials neural stem cells (NSCs) under different contexts remain unclear. Based on our generated single-nucleus atlas hippocampus across neonatal, adult, aging injury, we dissected molecular heterogeneity transcriptional dynamics hippocampal NSCs contexts. We further identified new specific neurogenic lineage markers that overcome lack specificity found some well-known markers. trajectory signatures, a subset exhibit quiescent properties after birth, most become deep quiescence during aging. Furthermore, certain are re-activated following stroke injury. Together, findings provide valuable insights into development, aging, re-activation NSCs, help to explain why is infrequently observed humans.
