ABSTRACT
An experimental autoimmune myocarditis rat model was established by subcutaneous injection of porcine myocardial myosin (PCM). The effect ET-1 receptor type B (ETBR) overexpression on observed via tail vein ETBR lentivirus in rats. We further investigated the mechanisms involved regulation overexpression.Six rats were randomly selected from 24 male Lewis as NC group, and remaining 18 injected with PCM Day 0 7, to establish (EAM) model. initially immunized divided into three groups: EAM ETBR-oe GFP group. On 21 after initial immunization rats, cardiac echocardiography serum brain natriuretic peptide (BNP) analysis performed evaluate function, tissue HE staining assess inflammatory infiltration score, mRNA expression IFN-γ, IL-12, IL-17 detected qRT-PCR. Subsequently, immunohistochemical detect localization ICAM-1 proteins, verified qRT-PCR western blotting methods. On immunization, left ventricular end-diastolic diameter (LVEDd), end-systolic (LVEDs), BNP concentrations increased hearts group compared (P < 0.01), ejection fraction (EF) fractional shortening (FS) decreased those normal control (NC) 0.01).
