ABSTRACT
Acetaminophen (N-acetyl-para-aminophenol, APAP) is the most widely used antipyretic and anti-inflammatory drug in world. It reported that APAP-induced liver damage accounts for about half of all failure patients Europe United States. Magnesium isoglycyrrhizinate (MI) fourth-generation glycyrrhizic acid preparation developed China. has anti-inflammatory, hepatocyte membrane protection, function recovery effects. This study aimed to investigate effect MI on alleviating injury explore potential mechanisms. C57 BL/6 J mice were assess efficacy by detecting ALT, AST, H&E TUNEL staining. Liver samples from saline, APAP, APAP combined with group selected transcriptomics analysis. significantly prevented elevation AST. Hepatocyte necrosis was alleviated when co-treated assay. There no differences total GSH levels or GSH/GSSG ration between group. Western Blot showed didn't affect protein CYP2E1 expression, mitochondrial p-JNK cytosolic Endo G. GO analysis mitochondria main target reducing injury. also upregulated expression TFAM, NRF-1, PGC-1β Sirt1. restored mRNA oxidative phosphorylation genes recovered fell after administration. In conclusion, promoting biogenesis.
