ABSTRACT
To investigate the molecular mechanism by which pirfenidone inhibits scar formation through TGF-β/Smad pathway.Cultured rabbit tenon fibroblasts (RTFs) were treated with different concentrations of to determine its initial active concentration and optimum for inhibiting RTF proliferation using CCK-8 assay. In RTFs at optimal concentrations, expressions TGF-β3, collagen I III examined both immunofluorescence assay Western blotting, their mRNA expression levels detected RT-PCR.The 0.1 mg/mL 0.27 mg/mL, respectively. two 24 h, blotting showed significantly lowered protein as compared those in control group (P < 0.05). The also after treatment 0.05).Pirfenidone concentration-dependently possibly down-regulating TGF-β3 pathway.
