ABSTRACT
To explore the mechanism that mediates effect of soybean isoflavones (SI) against cerebral ischemia/reperfusion (I/R) injury in light regulation regional blood flow (rCBF), ferroptosis, inflammatory response and blood-brain barrier (BBB) permeability.A total 120 male SD rats were equally randomized into sham-operated group (Sham group), I/R SI pretreatment (SI group). Focal was induced latter two groups using a modified monofilament occlusion technique, intraoperative changes real-time cortex monitored laser Doppler flowmeter (LDF). The postoperative pathological morphology ultrastructure neurons BBB observed with optical transmission electron microscopy. neurological deficits assessed, severities infarction, brain edema disruption quantified. contents Fe2+, GSH, MDA MPO ischemic penumbra determined spectrophotometric tests. Serum levels TNF-α IL-1βwere analyzed ELISA, expressions GPX4, MMP-9 occludin around lesion detected Western blotting immunohistochemistry.The rCBF sharply reduced after successful insertion monofilament. Compared those Sham group, showed significantly increased deficit scores, infarction volume, water content Evans blue permeability (P < 0.01), decreased Fe2+ level, GSH GPX4 expression serum IL-1β lowered 0.01). All these ameliorated pretreated IS prior to 0.05 or 0.01).SI preconditioning reduces possibly by improving rCBF, inhibiting ferroptosis protecting BBB.
