ABSTRACT
This study aims to investigate the pathogenesis of chronic heart failure based on ferroptosis-mediated oxidative stress and predict targets Shenfu Injection in treating failure. A rat model was established by isoproterenol induction method. According random number table method, modeled rats were assigned into three groups: a group, ferrostatin-1(ferroptosis inhibitor) group. In addition, normal group designed. After 15 days intervention, cardiac mass index left ventricular determined. Echocardiography employed eva-luate function. Hematoxylin-eosin staining Masson reveal pathological changes fibrosis heart, Prussian blue detect aggregation iron ions myocardial tissue. Transmission electron microscopy observe mitochondrion ultrastructure Colorimetry adopted measure levels metabolism, lipid peroxidation, antioxidant indicators. Flow cytometry content lipid-reactive oxygen species(ROS) fluorescence intensity ROS. Western blot RT-qPCR determine protein mRNA levels, respectively, ferroptosis-related factors The results showed that had reduced function, elevated total Fe~(2+), lowered level glutathione(GSH), increased malondialdehyde(MDA), decreased superoxide dismutase(SOD) glutathione peroxidase(GSH-Px), rising ROS lipid-ROS. fibrous tissue hyperplasia with inflammatory cell infiltration fibrosis, ion aggregation, characteristic mitochondrial specific for death. Moreover, showcased upregulated p53 COX2 downregulated GPX4, FTH1, SLC7A11, Nrf2 intervention significantly improved recovered indicators, deposition, structure alleviated fibrosis. Furthermore, can improve function regulating inhibiting ferroptosis, reducing injury.
