ABSTRACT
This study investigated the effect of Erchen Decoction(ECD) on prevention non-alcoholic steatohepatitis(NASH) in mice and explored its possible mechanism, so as to provide scientific data for clinical application ECD NASH. C57BL/6 male were randomly divided into normal group(methionine choline supplement, MCS), model deficient, MCD), low-dose group(ECD_L, 6 g·kg~(-1)), medium-dose group(ECD_M, 12 high-dose group(ECD_H, 24 with eight each group. The MCS group was fed an diet, other groups MCD diet. given corresponding diets, but drug intervention low-, medium-, ECD(10 mL·kg~(-1)·d~(-1)) by intragastric administration six weeks basis diet feeding, could eat drink freely during whole experiment. At end experiment, fasted overnight(12 h) anesthetized 20% urethane. Thereafter, blood liver tissue collected. serum used detect levels alanine aminotransferase(ALT), aspartate aminotransaminase(AST), interleukin-1β(IL-1β), interleukin-6(IL-6), interleukin-10(IL-10), tumor necrosis factor-α(TNF-α). Liver processed hematoxylin-eosin(HE) staining hepatic histological analysis detection expression genes proteins related nuclear factor erythroid 2-related 2/glutathione peroxidase 4(Nrf2/GPX4) pathway real-time quantitative reverse transcriptase-polymerase chain reaction(RT-qPCR) Western blot analysis, respectively. results showed that compared group, higher ALT AST levels; HE exhibited fat vacuoles obvious inflammatory cell infiltration tissue; IL-1β, IL-6, TNF-α significantly increased, IL-10 level decreased. mRNA expressions fatty acid synthase(FASN), monocyte chemoattractant protein-1(MCP-1), IL-1β up-regulated, while those GPX4, Nrf2, NAD(P)H:quinine oxidoreductase(NQO1) down-regulated. Compared ECD_M ECD_H decreased, ECD_L number degree improved; increased only FASN, MCP-1, down-regulated, GPX4 NQO1 up-regulated. Nrf2 protein after administration, FASN decreased; groups. In summary, can reduce lipid accumulation, oxidative stress, inflammation, injury NASH mice, which may be activation Nrf2/GPX4 pathway.
