ABSTRACT
Aconitum vilmorinianum is an authentic and superior medicinal herbal in Yunnan, which rich yunaconitine other diterpene alkaloids. Diterpene alkaloids are its main active components. Farnesyl pyrophosphate synthase(FPS) a key enzyme the terpene biosynthetic pathway plays important role alkaloid biosynthesis. Functional studies of FPS help to reveal molecular mechanism In this study, one gene(AvFPS) was selected based on transcriptome data A. vilmorinianum. Its full-length sequence cloned, bioinformatic analysis, functional verification, gene expression analysis were performed. The open reading frame(ORF) AvFPS 1 056 bp, encoding 351 amino acids. weight 41 kDa. had two typical conserved domains isopentenyl transferase, " DDIMD" DDYXD". recombinant protein expressed Escherichia coli, purified used for vitro enzymatic reaction. results revealed that able catalyze synthesis farnesyl pyrophosphate(FPP). qRT-PCR showed roots, stems, leaves, flowers vilmorinianum, with highest level roots. significantly up-regulated by MeJA induction. This study clarified catalytic function AvFPS, pattern different tissue, as well at time induced MeJA, provided reference deeper understanding biosynthesis diterpenoid
