ABSTRACT
This study aims to explore the mechanism of Zhuanggu Jianxi Decoction in reducing synovial tissue inflammation human knee osteoarthritis(KOA) via liver X receptors(LXRs)/nuclear factor(NF)-κB signaling pathway. The samples were collected from 5 healthy volunteers and 30 KOA synovitis patients cultured vitro. heathy set as normal group, those randomized into synovitis, Decoction, LXRα inhibitor, N-CoR inhibitor groups. groups treated with 10% blank serum, drug-containing serum+LXRα serum+N-CoR respectively, for 7 days. After intervention, each group stained hematoxylin-eosin observation scoring pathological changes. expression intensity fibroblast-specific marker α-smooth musle actin(α-SMA) was observed by immunofluorescence staining. levels interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α), matrix metalloproteinase-3(MMP-3), metalloproteinase-13(MMP-13) supernatant synovium homogenate determined ELISA. mRNA protein LXRα, N-CoR, P50, P65 RT-qPCR Western blot, respectively. results showed that compared showcased obvious lining cell proliferation, inflammatory infiltration, disarrangement, increased histopathological score(P<0.05), enhanced α-SMA fluorescence number fibroblasts(P<0.05), elevated IL-1β, TNF-α, MMP-3, MMP-13 tissue(P<0.05), down-regulated up-regulated P50 P65(P<0.05). Compared alleviated changes, declined score decreased lowered MMP-13(P<0.05), P65(P<0.05) tissue. aggravated risen above indicated could alleviate upregulating LXRs/NF-κB pathway downregulate reduce activity NF-κB
