ABSTRACT
To analyze the reasons for ABO blood group forward typing and reverse incompatibility of proband by serological molecular biological tests, so as to clarify genetic rules determine reasonable transfusion strategy.On basis testing results, PCR-SSP method was utilized exon sequencing in peripheral 5 family members, results were comprehensively analyzed.The incompatibility, test showed that there a c.700C>G mutation compared with B101, which consistent B(A)02 subtype, genotype B(A)02/O02. The elder brother same proband. nephew also detected c.700C>G, A102/B(A) 02.When result standard occurs, using biology detection technology explore is an effective confirm ensures safety clinical transfusion.B(A)血型鉴定及其临床输血策略研究.通过血清学及分子生物学检测分析1例先证者ABO血型正反不符的原因,以明确其血型及遗传规律并制定合理的输血策略.根据血清学结果,利用PCR-SSP方法对1例患者及其家庭5位成员的外周血进行ABO基因外显子测序,综合分析血型结果.先证者血型正反不符,分子生物学检测结果与B101对比多一个c.700C>G,符合B(A)02亚型,基因型为B(A)02/O02型;其哥哥检测结果同先证者;先证者侄子亦检测出c.700C>G,基因型为A102/B(A)02型.标准血型血清学鉴定ABO血型出现正反定型不符时,利用分子生物学检测技术发现突变点是证实ABO亚型的有效方法,为临床输血提供安全保障.
