ABSTRACT
Recent studies have shown that human bone marrow-derived mesenchymal stem cells (hBM-MSCs) several drawbacks in treating critical-sized defect (CSD). Secretome may offer considerable advantages over living terms of potency, manufacturing and storing easiness, potential as a ready-to-go osteoinductive agent. However, thus far, there are no regarding the efficacy secretome healing. The objective this study is to investigate effect rat models with CSD.This was an experimental post-test only control group design using 60 skeletally mature Sprague Dawley which divided evenly into 5 treatment groups (MSC only, MSC + Secretome, BMP-2, Control Normal Saline). We used Bone Marrow derived research. created by performing osteotomy treated according groups. Rats were sacrificed on 2nd 4th week we measured radiological outcome Radiographic Union Score for Tibia (RUST) histomorphometric (callus, osseous, cartilage, fibrous, void area) evaluation Image J.There difference weight rats between before after intervention. RUST score all intervention significantly higher than group, however, MSC-only not statistically significant group. There groups.Histomorphometric showed total callus formation widest MSC+Secretome+BMP-2 combination while osseous area found highest secretome-only group.Secretome, whether solely or combined BM-MSC novel, potent bone-healing agent CSD models.
