Effect of methylene blue pretreatment on acute kidney injury induced by sepsis in rats

ABSTRACT
Objective To evaluate the effect of methylene blue pertreatment on acute kidney injury induced by sepsis in rats.Methods Ninety male Sprague-Dawley rats,aged 1.5-2.5 months,weighing 200-250 g,were randomly assigned into 3 groups (n =30 each) using a random number table:sham operation group (S group),cecal ligation and puncture (CLP) (MB group).The rats were anesthetized with 10% chloral hydrate 350 mg/kg.Normal saline 0.8 ml was injected via caudal vein CLP groups,while 15 mg/kg (in normal ml) MB.Sepsis after end administration MB groups.Twenty animals each chosen observed for 72 h survival rate.Ten sacrificed at 18 specimens removed microscopic examination determination expression poly (ADP-ribose) polymerase (PARP-1) immunohistochemistry Western blot.Blood samples taken from heart serum concentrations blood urea nitrogen (BUN),creatinine (Cr),cystatin C neutrophil gelatinase-associated lipocalin (NGAL).Results Compared S,the rate significantly decreased 24,48 operation,and BUN,Cr,cystatin NGAL PARP-1 tissues increased (P < 0.05).Compared CLP,the 24 48 0.05).The pathological changes attenuated as compared CLP.Conclusion Methylene can attenuate through down-regulating PARP-1.
