ABSTRACT
Objective
To investigate the effect of paired related homoeobox 1 (Prrx1) on epithelial-mesenchymal transition (EMT) and migration ability breast cancer cells.
Methods
Breast cell lines (including BT-549, MDA-MB-468 MCF-7) were screened by real-time quantitative polymerase chain reaction (Real-time PCR) to obtain cells with lowest expression Prrx1. Then transfected Prrx1 over-expression letiviral vectors. The morphologic changes observed after transfection was verified be accomplished through Western blotting. At last, influence detected Transwell assay.
Results
The exprossioris mRNA in MCF-7, MDA-MB-468, BT-549 are 15.05±0.18, 11.78±0.42, 7.98±0.37. Thus, MCF-7 three kinds cells. over-expressing It found that morphology these changed from polygon long spindle appearance. Real-time PCR results showed experimental, control blank groups 2.16±0.07, 10.18±0.07 10.2±0.07, those E-adherin 4.00±0.02, 3.56±0.02 3.51±0.05, Vimentin 8.79±0.42, 10.18±0.06 9.97±0.05, respectively. levels Prrx1, vimentin 259.57, 0.71, 2.27 times compared without respectively (t=227.100, P=0.000; t=6.007, t=11.900, P=0.000), while difference between group lentiviral vectors not statistically significant (t=1.315, P=0.202; t=0.730, P=0.476; t=2.052, P=0.057). assay number migrating (73.40±10.21) greater than both non-treated (45.60±6.62) (44.60±8.24; t=6.837, (t=0.295, P=0.772).
Conclusion
Prrx1 can promote EMT improve their ability.
