ABSTRACT
Objective
To explore the feasibility of single micro extrusion 3D printing method for skin tissue.
Methods
Fibroblasts were isolated and cultured from human dermis identified by immunofluorescence staining. Fibroblasts HaCaT cells used as seed cells. Gelatin, hyaluronic acid fibrinogen to form scaffolds. HE staining observe cell distribution printed skin. The viability in tissue was detected living/dead kit. Toluidine blue penetration assay detect barrier function skin.
Results
More than 98% fibroblasts vimentin positive. When concentration epidermis 5 mg/ml 1 respectively, (4×106 cells/ml) 8 layers, (5×106 layer, prepared air-liquid interface 7 days detection showed that well stratified with thickness being (63.5±3.5) μm. (90.2±0.9)% (95.3±0.8)%, after submerge culture 2 days. After days, had similar normal skin.
Conclusion
We have established a tissue, successfully tissue-engineered structure, which may become an effective mass production future.
