A patient with recurrent multifocal glioblastoma received chimeric antigen receptor (CAR)-engineered T cells targeting the tumor-associated interleukin-13 alpha 2 (IL13Rα2). Multiple infusions of CAR were administered over 220 days through two intracranial delivery routes - into resected tumor cavity followed by ventricular system. Intracranial IL13Rα2-targeted not associated any toxic effects grade 3 or higher. After T-cell treatment, regression all and spinal tumors was observed, along...

Extracting sentiment from text is a hard semantic problem. We develop methodology for extracting small investor stock message boards. The algorithm comprises different classifier algorithms coupled together by voting scheme. Accuracy levels are similar to widely used Bayes classifiers, but false positives lower and accuracy higher. Time series cross-sectional aggregation of information improves the quality resultant index, particularly in presence slang ambiguity. Empirical applications...

Recent advances in small inexpensive sensors, low-power processing, and activity modeling have enabled applications that use on-body sensing machine learning to infer people's activities throughout everyday life. To address the growing rate of sedentary lifestyles, we developed a system, UbiFit Garden, which uses these technologies personal, mobile display encourage physical activity. We conducted 3-week field trial 12 participants used system report findings focusing on their experiences...

Traditional problem determination techniques rely on static dependency models that are difficult to generate accurately in today's large, distributed, and dynamic application environments such as e-commerce systems. We present a analysis methodology automates these by 1) coarse-grained tagging of numerous real client requests they travel through the system 2) using data mining correlate believed failures successes determine which components most likely be at fault. To validate our...

It is difficult to assign antibody specificity for highly sensitized patients using a cell panel with multiple antigens per reaction. We describe here single antigen bead accurate identification of human leukocyte (HLA) specificities by flow cytometry.A total 110 recombinant HLAs, including 34 A locus alleles, 57 B and 19 C were produced mammalian expression system. These coated onto eight different colored microbeads, which mixed together in one tube simultaneous detection HLA antibodies...

Chimeric antigen receptor T cell (CAR-T) therapy is an emerging strategy to improve treatment outcomes for recurrent high-grade glioma, a cancer that responds poorly current therapies. Here we report completed phase I trial evaluating IL-13Rα2-targeted CAR-T cells in 65 patients with the majority being glioblastoma (rGBM). Primary objectives were safety and feasibility, maximum tolerated dose/maximum feasible dose recommended 2 plan. Secondary included overall survival, disease response,...

Abstract Polypharmacology drugs—compounds that inhibit multiple proteins—have many applications but are difficult to design. To address this challenge we have developed POLYGON, an approach polypharmacology based on generative reinforcement learning. POLYGON embeds chemical space and iteratively samples it generate new molecular structures; these rewarded by the predicted ability each of two protein targets drug-likeness ease-of-synthesis. In binding data for >100,000 compounds, correctly...

Transactional Coherence and Consistency (TCC) offers a way to simplify parallel programming by executing all code within transactions. In TCC systems, transactions serve as the fundamental unit of work, communication coherence. As each transaction completes, it writes its newly produced state shared memory atomically, while restarting other processors that have speculatively read stale data. With this mechanism, TCC-based system automatically handles data synchronization correctly, without...
