Efficient production of bispecific antibodies (BsAbs) in single mammalian cells is essential for basic research and industrial manufacturing. However, preventing unwanted pairing heavy chains (HCs) light (LCs) a challenging task. To address this, we created an engineering technology preferential cognate HC/LC HC/HC paring called FAST-Ig (Four-chain Assembly by electrostatic Steering Technology – Immunoglobulin), applied it to NXT007, BsAb the treatment hemophilia A. We introduced charged...
Abstract: We studied the serum lipoprotein and apolipoprotein profiles in 44 patients with sporadic late‐onset Alzheimer's dementia 43 vascular dementia. The levels of high‐density (HDL) cholesterol were lower both patient groups than a control group. Apolipoprotein A I II have decreased groups, especially HDL‐cholesterol correlated positively level I, but not II. ratios I/A increased groups. disproportionally may involve pathological process senile
Summary The RNA-guided endonuclease Cpf1 is a promising tool for genome editing in eukaryotic cells 1-5 . Compared to other platforms, offers distinct advantages, such as the ability easily target multiple genes simultaneously 3 , well low rates of off-target activity 4, 5 However, Acidaminococcus sp. BV3L6 (AsCpf1), which has been successfully harnessed editing, can only robustly cleave sites preceded by TTTV protospacer adjacent motif (PAM), may limit its practical utility. To address this...
The CRISPR-associated endonuclease Cas9 can be targeted to specific genomic loci by single guide RNAs (sgRNAs).We first solved the crystal structure of Streptococcus pyrogenes (SpCas9) in complex with sgRNA and its target DNA at 2.5 A resolution. The revealed a bilobed architecture consisting recognition nuclease lobes (Rec Nuc lobes, respectively), accommodating sgRNA:DNA heteroduplex positively-charged groove their interface. While Rec lobe is essential for binding DNA, contains HNH RuvC...
Immunohistochemical examinations using monoclonal antibody against subunit IV of cytochrome c oxidase were carried out in the muscle patients with various mitochondrial myopathies having histochemical partial deficiency activity. The ragged red fibers showed positive reaction for even deficient fibers. However, normal histological appearance, immunohistochemical varied. While all reactivity had no detectable activity, protein was not cause
