The aim of this study was to test the Brilliant Cresyl Blue (BCB) stain select prepubertal sheep oocytes for in vitro blastocyst production. Oocyte diameter, mitochondrial activity, maturation-promoting factor (MPF) activity and mRNA relative expression (RE) genes related metabolism (ATPase Na(+)/K(+) transporting α 1 (ATP1A1) cytochrome c oxidase subunit (COX1)) constitutive function cell (cytoplasmic polyadenylation-element-binding protein (CPEB) S100A10) were assessed. Immature exposed...

Melatonin is a universal antioxidant that improves in vitro embryo production several species. The aims of this study were to determine the melatonin concentration ovarian follicular fluid (FF) juvenile goats and effect during maturation (IVM) on development. FF was 0.57--1.07×10-9 M, increasing with diameter. Oocytes matured, fertilised cultured under conventional conditions. Blastocyst development, quality levels reactive oxygen species (ROS) reduced glutathione assessed. In Experiment 1...

Melatonin enhances in vitro embryo development several species by improving the oocyte developmental competence during maturation (IVM). has a wide range of actions, from scavenging reactive oxygen (ROS) to regulating gene expression, and it can also act way melatonin receptors. The aim this study was determine mechanism action IVM juvenile goat oocytes role membrane receptor 1 immunolocalized cumulus cells before after 24 hr IVM. effect on tested three experimental groups: (a) control, (b)...

Contents The aim of this study was to test the effect insulin–transferrin–selenium (ITS) and L‐ascorbic acid (AA) supplementation hormonal level during in vitro maturation (IVM) small oocytes from pre‐pubertal goat on blastocyst yield quality. Concretely, we used four media: conventional IVM medium (CM), growth (GM: CM+ITS+AA low hormones), modified CM (mCM: with hormones) GM (mGM: normal hormones). Cumulus–oocyte complexes (COCs) were classified into two categories according oocyte...

In goats, embryo oocyte competence is affected by follicle size regardless the age of females. previous studies we have found differences in blastocyst development between oocytes coming small (<3 mm) and large follicles (>3 prepubertal (1−2 months-old) goats. Oocyte Follicular Fluid (FF) composition changes throughout growth. The aim this study was to analyze Fatty Acids (FAs) metabolomic profiles FF recovered from goats adult FAs were analyzed chromatography metabolites 1H-Nuclear Magnetic...

A method for the determination of trace amounts Pd, Fe and Cu in pure gold has been established using inductively coupled plasma atomic emission spectrometry (ICP-AES). The most suitable spectral lines analysis have selected by considering two essential criteria: sensitivity interferences. samples were dissolved with aqua regia determinations carried out direct calibration synthetic standards containing regia. Precision detection limits investigated. inhomogeneity sample was inferred from a...

In our laboratory, we have been working for more than 20 years on in vitro embryo production from prepubertal goat oocytes. We observed significant differences blastocyst according to the season of year. The present study is aimed asses these summer, autumn, winter, and spring. Abstract A method for the direct determination of Cd in soil extracts by graphite furnace atomic absorption spectrosopy (GFAAS) has been developed. Optimum operating conditions, analyte modifiers and matrix interferences have investigated. Characteristic amounts are 0.34pg siliceous samples 0.45 pg calcareous soils. The precision based on ten replicate readings standards, was 1.6%. determined proposed methods results compared with those obtained ICP technique.
