A synthetic luciferin comprising an imidazopyrazinone core, named HuLumino1, was designed to generate specific bioluminescence with human serum albumin (HSA) in real samples. HuLumino1 developed by attaching a methoxy-terminated alkyl chain C-6 of coelenterazine and eliminating benzyl group at C-8. HSA levels were quantified within 5% error margins enzyme-linked immunosorbent assay without the need for any sample pretreatments because high specificity HuLumino1.

Fibroblast growth factor 21 (FGF21) is an effective metabolic regulator of glucose and lipid homeostasis in the context insulin resistance, intolerance dyslipidemia diabetic rodents monkeys, peroxisome proliferator-activated receptor α (PPARα) directly induces FGF21 expression rodent liver. Recent findings suggest that effects regulation qualitatively differ between humans. Here, we examined PPARα PPARγ agonists on mRNA mouse liver cultured hepatocytes. Intraperitoneal injection both...

The circadian clock is a cell-autonomous endogenous system that generates rhythms in the behavior and physiology of most organisms. We previously reported harmala alkaloid, harmine, lengthens period Bmal1 transcription NIH 3T3 fibroblasts. Clock protein dynamics were examined using real-time reporter assays PER2::LUC to determine effects harmine on central suprachiasmatic nucleus (SCN). Harmine significantly lengthened expression embryonic fibroblasts, neuronal cells differentiated from...

We have been investigating transcriptional regulation of the BMAL1 gene, a critical component mammalian clock system including DNA methylation. Here, more detailed analysis methylation proceeded in RPMI8402 lymphoma cells. found that CpG islands and PER2 promoters were hyper- hypomethylated, respectively 5-aza-2'-deoxycytidine (aza-dC) not only enhanced gene expression but also oscillation within 24 h That is, such hypermethylation promoter restricted which was recovered by aza-dC 1 day...

Abstract The control of the circadian rhythm is important for health because it regulates physiological functions and associated with hazards. We aimed to identify a biomarker status in human saliva, since collecting saliva non-invasive, straightforward, cost-effective. Among 500 genes potentially controlled by salivary clock identified using chromatin immunoprecipitation (ChIP) assays, 22 them showed reasonable transcriptional responses according DNA array model system. these genes, ARRB1,...

Withanolide derivatives have anticancer, anti-inflammatory, and other functions are components of Indian traditional Ayurvedic medicine. Here, we found that 2,3-dihydro-3β-methoxy withaferin-A (3βmWi-A), a derivative (Wi-A) belonging to class withanolides abundant in Ashwagandha (Withania somnifera), lengthened the period circadian clock. This compound dose-dependently elongated rhythms Sarcoma 180 cancer cells normal fibroblasts including NIH3T3 spontaneously immortalized mouse embryonic...

Abstract:: Circadian rhythms of innate 24 h cycles comprise well-conserved biological phenomena from cyanobacteria to mammalian. They are driven by light and regulated clock genes that work as transcription factors control the expression many other physiological functions in cells. The ~ 40% protein-coding shows oscillation patterns mice, implying their importance normal body functions. Indeed, behavioural rhythmicity generated through genes-mediated multiple mechanisms affects quality life...

We genetically manipulated HaCaT cells, a spontaneously immortalised normal keratinocyte cell line, to stably express two different coloured luciferase reporter genes, driven by interleukin 8 ( IL‐8 ) and ubiquitin‐C UBC promoters, respectively. Subsequently, we generated three‐dimensional (3D) skin‐like in vitro composite (SLIC) utilising these with the objective of monitoring bioluminescence emitted from SLIC. This SLIC was on non‐woven silica fibre membranes differentiation medium....

We have developed a comprehensive method for analyzing gene expression levels by combining the fluorous-fluorescence hybrid probes, oligonucleic acids modified with perfluoroalkyl tag and fluorescent fluorous liquid chromatography (LC) – fluorescence detection. When Taq polymerase chain reaction (PCR) was performed using this probe, mononucleotides were released according to amount of template targeted. Because each probe differs in linker length type nucleobase, multiple can be analyzed LC...
