The biophysical properties and physiologic effects of a series clinical exogenous pulmonary surfactants was compared to determine the importance hydrophobic apoproteins (SP-B C) as constituents these preparations. three studied, calf lung surfactant extract (CLSE), Survanta (Surfactant-TA), Exosurf, all contain dipalmitoyl phosphatidylcholine (DPPC) their major constituent. CLSE also 1 2% SP-B,C but Exosurf has additives hexadecanol tyloxapol instead enhance activity DPPC. In adsorption...

Surface pressure-area (*-A) isotherms during dynamic cycling were measured for films of dipalmitoyl phosphatidylcholine (DPPC) and column-separated fractions calf lung surfactant extract (CLSE).Emphasis was on defining the relative importance phospholipids (PPL), neutral lipids (N), hydrophobic proteins (SP) in facilitating respreading surface tension lowering within interfacial film itself.Solvent-spread a Wilhelmy balance studied at 23' 37OC over range rates initial concentrations giving...

This paper reports the detailed composition of molecular species phosphatidylcholines (PCs) in pulmonary surfactant from calves. PC isolated by thin-layer chromatography (TLC) was converted to benzoylated diradyl glyceride derivatives, which were separated TLC according linkage group. Quantification groups analysis total fatty acid content demonstrated that contained 97.2% diacyl, 2.4% alkyl-acyl, and 0.4% alkenyl-acyl compounds. The diacyl alkyl-acyl diglyceride derivatives into individual...

This paper examines two factors, shape deformation and surface viscosity, that affect measurements of tension lung surfactants with the oscillating bubble surfactometer. At lower tensions, compressed in this apparatus becomes deformed to an oblate ellipsoid cannot be analyzed rigorously using simplified (spherical) Laplace equation calculate from interfacial pressure drop. However, for small air bubbles present apparatus, analysis more general equations ellipsoids revolution shows effects...

We have shown previously that lateral compression of pulmonary surfactant monolayers initially induces separation two phases but these remix when the films become more dense (1). In studies reported here, we used fluorescence microscopy to examine role different constituents in remixing separated phases. Subfractions containing only purified phospholipids (PPL), proteins and (SP&PL), neutral (N&PL) were obtained by chromatographic components extracted calf (calf lung extract, CLSE)....

The inhibitory effects of oleic acid (OA) on the surface activity pulmonary surfactant were characterized by use oscillating bubble surfactometer, Wilhelmy balance, and excised rat lungs. Oscillating studies showed that OA prevented lavaged calf [0.5 mM phospholipid (PL)] from lowering tension below 15 mN/m at or above a molar ratio OA/PL = 0.5. In contrast to inhibition plasma proteins, increasing concentration did not eliminate acid, which occurred greater than 0.67 even concentrations 1.5...
