ABSTRACT Phagocytic cells form the first line of defense against infections by human fungal pathogen Candida albicans . Recent in vitro gene expression data suggest that upon phagocytosis macrophages, C. reprograms its metabolism to convert fatty acids into glucose inducing enzymes glyoxylate cycle and acid β-oxidation pathway. Here, we asked whether β-oxidation, a metabolic pathway localized peroxisomes, is essential for virulence constructing two double deletion strains: pex5Δ/pex5Δ...

Summary Previously, we have shown that PDE2 is required for hyphal development and cell wall integrity in Candida albicans. In the present study, investigated effects of its deletion by genome‐wide transcriptome profiling. Changes expression levels genes involved metabolism, transcription, protein nucleic acids synthesis, as well stress responses, membrane biogenesis, adherence virulence been observed. By comparing these changes with previously reported profiles pde2 Δ mutants Saccharomyces...

The glyoxylate cycle, a metabolic pathway required for generating C4 units from C2 compounds, is an important factor in virulence, both animal and plant pathogens. Here, we report the localization of key enzymes this isocitrate lyase (Icl1; EC 4.1.3.1) malate synthase (Mls1; 2.3.3.9), human fungal pathogen Candida albicans. Immunocytochemistry combination with subcellular fractionation showed that Icl1 Mls1 are localized to peroxisomes, independent carbon source used. Although lack consensus...

A Klebsiella pneumoniae epidemic strain that coproduced carbapenemase KPC-2 (K. 2) and 16S rRNA methylase ArmA has emerged in Poland. Four nonduplicate isolates from patients a hospital Warsaw, Poland, were found to carry the bla(KPC-2) armA genes on ca. 50-kb 90-kb plasmids, respectively. Tn4401 with 100-bp deletion variable region was detected all isolates. XbaI pulsed-field gel electrophoresis (PFGE) revealed 93.2% similarity of All resistant carbapenems 4,6-disubstituted 2-deoxystreptamines.

Salmonellosis remains the second most common zoonosis in Europe. Resistance to fluoroquinolones (FQs) Salmonella has been increasing worldwide, with WHO considering FQ-resistant spp. as high-priority pathogens. The aim of this study was a retrospective analysis molecular mechanisms FQ resistance, detected among clinical ciprofloxacin-resistant enterica belonging serotypes. whole genome sequences (WGS) tested isolates were also analysed for occurrence other antimicrobial resistance...

We characterized 17 clinical isolates of Klebsiella pneumoniae producing 16S rRNA methylase ArmA. The originated in Poland from 2002 to May 2010 and encompassed four XbaI-PFGE clusters. All the were resistant amikacin, gentamicin kanamycin (MIC range: 256-1024 mg l(-1)) carried armA gene on a large plasmid approximately 90 or 130 kb 15 2 isolates, respectively. was found ~10 ClaI restriction fragment flanked by same elements as Tn1548. bla(CTX-M) for CTX-M-type extended-spectrum β-lactamase....

Aminoglycosides are a group of antimicrobial agents still the most commonly used in treatment life-threatening bacterial infections human and animals. The emergence spread 16S rRNA methylases, which confer high-level resistance to majority clinically relevant aminoglycosides, constitute major public health concern. Our goal was evaluate distribution methylases among different species Enterobacteriaceae during five month-long survey tertiary hospital Warszawa, Poland. In survey, total 1770...

Abstract Background Transrectal ultrasound-guided prostate biopsy (TRUS-Bx) is considered an essential urological procedure for the histological diagnosis of cancer. It is, however, a “contaminated” which may lead to infectious complications. Recent studies suggest significant share fluoroquinolone-resistant rectal flora in post-biopsy infections. Methods The molecular mechanisms fluoroquinolone resistance, including PMQR (plasmid-mediated quinolone resistance) as well mutation QRDRs...
