The Commander complex is required for endosomal recycling of diverse transmembrane cargos and mutated in Ritscher-Schinzel syndrome. It comprises two sub-assemblies: Retriever composed VPS35L, VPS26C, VPS29; the CCC which contains twelve subunits: COMMD1-COMMD10 coiled-coil domain-containing (CCDC) proteins CCDC22 CCDC93. Combining X-ray crystallography, electron cryomicroscopy, silico predictions, we have assembled a complete structural model Commander. distantly related to Retromer but has...
Summary In cyanobacteria and chloroplasts, exposure to HL damages the photosynthetic apparatus, especially D 1 subunit of P hotosystem II . To avoid chronic photoinhibition, a PSII repair cycle operates replace damaged subunits with newly synthesised versions. determine sub‐cellular location this process, we examined localisation F ts H metalloproteases, some which are directly involved in degrading 1. We generated transformants cyanobacterium S ynechocystis sp. PCC 6803 expressing GFP...
This work examined the long-term effects of periodic high light stress on photosynthesis, morphology, and productivity low-light-acclimated Arabidopsis plants. Significant photoinhibition seedlings grown under low (100 μmol photons m−2 s−1) was observed at beginning treatment (three times a day for 30 min 1800 s−1). However, after 2 weeks treatment, similar photosynthesis yields (Fv/Fm) to those control plants were attained. The daily levels photochemical quenching measured in dark (qPd)...
Novel macrocyclic peptides are found that bind and modulate the function of retromer membrane trafficking complex.
Background In cyanobacteria the photosystems are localised to, and maintained in, specialist membranes called thylakoids. The mechanism driving biogenesis of thylakoid is still an open question, with only two potential factors, Vipp1 Alb3 currently identified. Methodology/Principal Findings We generated a slr1768 knockout using pGEM T-easy vector REDIRECT. By comparing growth pigment content (chlorophyll fluoresence) Δslr1768 mutant wild-type, we found that has conditional phenotype;...
ABSTRACT The Retromer complex (Vps35-Vps26-Vps29) is essential for endosomal membrane trafficking and signalling. Mutations in cause late-onset Parkinson’s disease, while viral bacterial pathogens can hijack the during cellular infection. To modulate probe its function we have created a novel series of macrocyclic peptides that bind with high affinity specificity. Crystal structures show majority cyclic to Vps29 via Pro-Leu-containing sequence, structurally mimicking known interactors such...
Abstract The sorting nexin SNX17 controls endosome-to-cell surface recycling of diverse transmembrane cargo proteins including integrins, the amyloid precursor protein and lipoprotein receptors. This requires association with multi-subunit Commander trafficking complex, which depends on C-terminus through unknown mechanisms. Using affinity enrichment proteomics, we find that a C-terminal peptide is not only sufficient for interaction but also associates members actin-associated PDZ LIM...

Abstract Co-immunoprecipitation of proteins coupled to mass spectrometry is critical for the understanding protein interaction networks. In instances where a suitable antibody not available, it common graft synthetic tags onto target sequences and allowing use commercially available antibodies affinity purification. A approach through FLAG-Tag co-immunoprecipitation. To allow selective elution complexes, competitive displacement using large molar excess tag peptides often carried out. Yet,...
