In bacteria, OxyR is a peroxide sensor and transcription regulator, which can sense the presence of reactive oxygen species induce antioxidant system. When cells are exposed to H2O2, protein activated via formation disulfide bond between two conserved cysteine residues (C199 C208). Deinococcus radiodurans, previously unreported special characteristic DrOxyR (DR0615) found with only one cysteine. dr0615 gene mutant hypersensitive but little ionizing radiation. Summary Two‐component systems are predominant signal transduction pathways in prokaryotes, and also exist many archaea as well some eukaryotes. A typical TCS consists of a histidine kinase cognate response regulator. In this study, novel gene encoding regulator (we designate it drRRA ) is identified to be essential for the extreme radioresistance Deinococcus radiodurans . DrRRA null mutant MR) sensitive gamma‐radiation compared with wild‐type strain. Transcriptional assays show that numerous...

Preliminary findings indicate that PprI is a regulatory protein stimulates transcription and translation of recA other DNA repair genes in response to damage the extremely radioresistant bacterium Deinococcus radiodurans. To define repertoire proteins regulated by investigate vivo mechanism gamma radiation, we performed comparative proteomics analyses on wild type (R1) pprI knock-out strain (YR1) under conditions ionizing irradiation. Results two-dimensional electrophoresis MALDI-TOF MS or...

The supramolecular assembly of a novel cyanine dye, 3,3′-di(3-sulfopropyl)-4,5,4′,5′-dibenzo-9-ethyl-thiacarbocyanine triethylammonium salt (ETC) was designed to verify specific intramolecular G-quadruplexes from duplex and single-strand DNAs. Spectral results have shown that ETC presented two major distinct signatures with in vitro: (i) dramatic changes the absorption spectra (including disappearance peak around 660 nm appearance independent new 584 nm); (ii) ∼70 times enhancement...

Sasa argenteastriatus, with abundant active compounds and high antioxidant activity in leaves, is a new leafy bamboo grove suitable for exploitation. To utilize it more effectively scientifically, we investigate the seasonal variations of composition its leaves activity. The argenteastriatus were collected on 5th day each month three same-sized sample plots from May 2009 to 2011. total flavonoids (TF): phenolics (TP) triterpenoid (TT) extracted contents analyzed by UV-spectrophotometer. A G-quadruplex is identified which can exhibit two different motifs, respectively, corresponding to Na+ and K+. The relative amount of each motif related the Na+/K+ ratio. Based on selective recognition motifs by a cyanine dye aggregate, method for both colorimetric quantitative measurements ratios constructed.

Abstract Background Deinococcus radiodurans accumulates high levels of manganese ions, and this is believed to be correlated with the radiation resistance ability microorganism. However, maintenance ion homeostasis in D. remains investigated. Results In study, we identified efflux protein (MntE) . The null mutant mntE was more sensitive than wild-type strain growth delayed manganese-supplemented media. Furthermore, there a substantial increase vivo concentration ions. Consistent these...

Double-minded supramolecules: The interactions between human serum albumin (HSA) and J-aggregation of a cyanine dye (PTC) in phosphate buffer solution are studied. In the presence HSA, achiral J-aggregate shows chirality. chirality can be inversed by adjusting ratio HSA to PTC pH value (see picture; CD=circular dichroism). Here we present direct evidence for the vital role of RecO in Deinococcus radiodurans's radioresistance. A recO null mutant was constructed using a deletion replacement method. The exhibited growth defect and extreme sensitivity to irradiation with gamma rays UV light. These results suggest that DNA repair this organism occurs mainly via RecF pathway.
