Ceramides are key intermediates in sphingolipid biosynthesis and potent signaling molecules. However, excess ceramide is toxic, causing growth arrest apoptosis. In this study, we identify a novel mechanism by which cells prevent the toxic accumulation of ceramides; they facilitate nonvesicular transfer from endoplasmic reticulum (ER) to Golgi complex, where ceramides converted complex sphingolipids. We find that yeast protein Nvj2p promotes ER complex. The tether generates close contacts...

Recent studies have reported intrinsic metabolic reprogramming in Pkd1 knock-out cells, implicating dysregulated cellular metabolism the pathogenesis of polycystic kidney disease. However, exact nature changes and their underlying cause remains controversial. We show herein that k o /ko renal epithelial cells impaired fatty acid utilization, abnormal mitochondrial morphology function, mitochondria kidneys ADPKD patients morphological alterations. further a C-terminal cleavage product...

Close contacts between organelles, often called membrane contact sites (MCSs), are regions where lipids exchanged organelles. Here, we identify a novel mechanism by which cells promote phospholipid exchange at MCSs. Previous studies have shown that phosphatidylserine (PS) synthase activity is highly enriched in portions of the endoplasmic reticulum (ER) with mitochondria. The objective this study was to determine whether enrichment promotes PS transport out ER. We found mitochondria more...

Three Bo beta fruct cDNAs encoding acid invertases were cloned from shoots of the green bamboo Bambusa oldhamii. On basis amino sequences their products and phylogenetic analyses, fruct1 fruct2 determined to encode cell wall invertases, whereas fruct3encodes a vacuolar invertase. The recombinant proteins encoded by fruct3 produced in Pichia pastoris purified near homogeneity using ammonium sulfate fractionation immobilized metal affinity chromatography. pH optima, pI values, substrate...

Ceramide is key intermediate for synthesis of sphingolipids, It also an important second messenger in signal transduction. However, overproduction ceramide triggers apoptosis and causes cell death. Our goal to understand how adapt itself from toxicity stress. synthesized ER, but transferred Golgi form complex sphingolipids. Such lipid trafficking events can be divided into vesicular non‐vesicular transport. Non‐vesicular transport usually occurs at Membrane contact sites (MCSs), which are...

Mitochondrial membrane biogenesis requires phospholipid import into mitochondria. One lipid that is critical for mitochondrial function, phosphatidylethanolamine (PE), synthesized in mitochondria from phosphatidylserine (PS) made the endoplasmic reticulum (ER) and imported PS transfer to occurs at regions of close contact between ER mitochondria, often called sites (MCSs). We investigated mechanism transport. It has long been known synthase activity enriched portions are associated with...
