A method to extract peptides and low molecular weight proteins from serum under denaturing conditions using acetonitrile containing 0.1% trifluoroacetic acid has been developed. The extraction procedure precipitates large, abundant simplify subsequent mass spectral analysis. This sample preparation provides an efficient way peptides, enabling them be compared identified different spectrometry approaches. Surface-enhanced laser desorption/ionization-time of flight analysis mouse blood samples...
Carbohydrate-protein conjugates are utilized extensively in basic research and as immunogens a variety of bacterial vaccines cancer vaccines. As result, there have been significant efforts to develop simple reliable methods for the construction these conjugates. While direct coupling via reductive amination is an appealing approach, reaction typically very inefficient. In this paper, we report improved conditions providing approximately 500% increase yield. addition optimizing series...
A coating that embodies omnidirectional optical properties over a wide range of wavelengths and possessing specific wetting capabilities is established on glass platforms.
The mutant β1,4-galactosyltransferase (β4Gal-T1), β4Gal-T1-Y289L, in contrast to wild-type β4Gal-T1, can transfer GalNAc from the sugar donor UDP-GalNAc acceptor, GlcNAc, with efficiency as good that of galactose UDP-Gal. Furthermore, also a modified sugar, C2 keto galactose, its UDP derivative O-GlcNAc modification on proteins provided functional handle for developing highly sensitive chemoenzymatic method detecting post-translational proteins. We report herein be transferred free GlcNAc...
We describe the formation and properties of atomically bonded, optical quality, nanostructured thin glass film coatings on plates, utilizing phase separation by spinodal decomposition in a sodium borosilicate system. Following deposition via magnetron sputtering, thermal processing differential etching, these are structurally superhydrophilic (i.e., display anti-fogging functionality) demonstrate robust mechanical superior abrasion resistance. After appropriate chemical surface modification,...
The authors investigate the properties of water drops on uniform arrays vertically aligned nanocones. A simple model predicts a variety behaviors depending nanocone aspect ratio. At low ratios, surface tension pulls into array, resulting in wetted drop with high contact angle hysteresis. effect is reversed, little liquid-solid and which easily slide off surface. Experiments show that transition occurs as gradual change angles, but an abrupt sliding angles.
Since the discovery of potentially mutagenic nitrosamine impurities in pharmaceuticals 2018, liquid chromatography-mass spectrometry (LC-MS) methods have been widely applied for trace level quantification nitrosamines. As more nitrosamines and drug samples are analyzed, method accuracy is increasingly recognized as a major analytical challenge. Matrix effects can significant adverse impact on but not attracted much attention literature. In this study, we observed severe matrix when adapting...
By exploiting phase-separation in oxide materials, we present a simple and potentially low-cost approach to create exceptional superhydrophobicity thin-film based coatings. selecting the TiO2–Cu2O system depositing through magnetron sputtering onto single crystal metal templates, demonstrate growth of nanostructured, chemically phase-segregated composite films. These coatings, after appropriate chemical surface modification, robust, non-wetting Cassie–Baxter state yield an superhydrophobic...
Pentafluorobenzyl chloroformate (PFB-chloroformate) has been utilized as a derivatization reagent to impart electron affinity and provide structurally relevant fragmentation in capture negative ion chemical ionization mass spectrometry (ECNICI-MS). Phenylalanine (Phe) decanol were used model analytes. The conditions for their the chromatographic spectrometric properties of derivatives are reported. aqueous solution was derivatized one step by using PFB-chloroformate mixture water, ethanol,...
Abstract Since its introduction as an ionization technique in mass spectrometry, matrix‐assisted laser desorption (MALDI) has been applied to a wide range of applications. Quantitative small molecule analysis by MALDI, however, is limited due the presence intense signals from matrix coupled with non‐homogeneous surfaces. The surface used nano‐structured (NALDI) eliminates need for and resulting interferences, allows quantitative molecules. This study was designed analyze quantitate...
We report a novel method of producing ordered arrays glass nanocones with precisely controlled height, lattice constant and aspect ratio. As nanochannel glass, fibre drawing, bundling redrawing are used to produce structured composite material. The surface the is etched form through differential etching process. ranges from 40 µm 1.6 µm, while ratio varied 0.4 13 by simple changes in chemistry hydrofluoric acid solution.
The Student Nitric Oxide Explorer (SNOE) is a small scientific spacecraft designed to launch on PegasusTM XL vehicle for the Demonstration Initiative. Its goals are measure nitric oxide density in lower thermosphere and analyze solar magnetospheric influences that create it cause its abundance vary dramatically. SNOE ('snowy') instrumentation being built at University of Colorado Laboratory Atmospheric Space Physics (LASP) by team scientists, engineers, students. compact hexagonal...
To date, most of the proteomic analyses on lung cancer tissue samples have been performed using surgical specimens, which are obtained after a diagnosis is made. determine if signature from bronchoscopic biopsy could be found to assist with diagnosis, 50 and 13 adjacent normal were analyzed histology‐directed, matrix‐assisted laser desorption/ionization (MALDI) mass spectrometry (MS). Lung cryosectioned, sinapinic acid was robotically deposited areas each section enriched in epithelial...
The oxidation of naphthalene-1,2-dihydrodiol (ND) to o-naphthoquinone (NQ) in the lens is believed be responsible for formation cataracts naphthalene-fed rats. Studies using either recombinant rat (RLAR) or human muscle aldose reductase (HMAR) incubated vitro with ND presence NAD(P) verified that (EC 1.1.1.21) dihydrodiol dehydrogenase catalyzes NQ. Kinetic studies Vmax/Km indicated RLAR NAD-dependent an optimal pH 9.0. corresponding activity HMAR was lower than enzyme. metabolite produced...
