Mutations in the gene encoding Cu/Zn superoxide dismutase-1 cause amyotrophic lateral sclerosis. Superoxide mutations decrease protein stability and promote aggregation. The mutant monomer is thought to be an intermediate pathway from dimer aggregate. Here we find that monomeric copper-apo, zinc-holo structurally perturbed apo-protein aggregates without reattainment of monomer–dimer equilibrium. Intervention stabilize inhibit aggregation regarded as a potential therapeutic strategy. We...

Opening of the mitochondrial permeability transition pore (MPTP) causes dysfunction and necrosis in acute pancreatitis (AP), a condition without specific drug treatment. Cyclophilin D (CypD) is matrix peptidyl-prolyl isomerase that regulates MPTP target for AP. We have synthesized urea-based small molecule inhibitors cyclophilins tested them against CypD using binding activity assays. Thermodynamic profiles CypD/inhibitor interactions were determined by isothermal titration calorimetry....

Closing the deal: A ring-closing metathesis (RCM)/aromatization protocol, followed by a regioselective Negishi cross-coupling reaction and macrolactonization, with subsequent RCM mediate synthesis of (−)-(Z)-deoxypukalide in 12 linear steps 15 % overall yield (see retrosynthesis; TBS=tert-butyldimethylsilyl, TIPS=triisopropylsilyl). Carboxyl methyltransferase (CMT) enzymes catalyse the biomethylation of carboxylic acids under aqueous conditions and have potential for use in synthetic enzyme cascades. Herein we report that FtpM from Aspergillus fumigatus can methylate a broad range aromatic mono- dicarboxylic good to excellent conversions. The shows high regioselectivity on its natural substrate fumaryl-l-tyrosine, trans, trans-muconic acid number tested. Dicarboxylic are generally better substrates than monocarboxylic...

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder characterised by the selective dysfunction and death of upper lower motor neurons. Median survival rates are between 3 5 years after diagnosis. Mutations in gene encoding Cu/Zn superoxide dismutase (SOD1) have been linked to subset familial forms ALS (fALS). Herein, we describe fragment- based drug discovery (FBDD) approach for investigation small molecule binding sites SOD1. X-ray crystallography has used as...

Lithium metal reductions are widely employed in organic synthesis, where it is common to employ a "mediator" speed up the electron transfer kinetics. We present experimental data for electrode kinetics of reduction most mediator, 4,4'-di-tert-butyl-1,1'-biphenyl (DBB) tetrahydrofuran (THF) over range temperatures. Using corresponding oxidation lithium we quantitative estimates kinetic advantage use DBB as mediator reductions, over, particular, direct using metal.

Abstract The reduction of methyl benzoate was studied in tetrahydrofuran (THF), 0.1 M TBAP, both at platinum and glassy carbon macroelectrodes microelectrodes. While cyclic voltammograms obtained show extensive distortion due to ohmic drop, this is negligible for system benzoate/methyl radical anion found be chemically reversible. fitting chronoamperometric measurements using the Shoup Szabo's expression microdisc electrodes allowed us estimate diffusion coefficient benzoate. Cyclic...

Sulfotransferases (STs) catalyse the transfer of a sulfonyl group ('sulfation') from enzyme co-factor 3'-phosphoadenosine 5'-phosphosulfate (PAPS) to variety biomolecules. Tyrosine sulfation proteins and carbohydrate play crucial role in many protein-protein interactions cell signalling pathways extracellular matrix. This is catalysed by several membrane-bound STs, including tyrosylprotein sulfotransferase 1 (TPST1) heparan sulfate 2-O-sulfotransferase (HS2ST1). Recently, involvement these...

Electrochemical benefit from a large accessible potential window. In practice, it is however necessary to avoid presence humidity cell.
