We constructed five chimeric clones between human immunodeficiency virus type 1 (HIV-1) and simian (SIVMAC) four SIVMAC mutants by recombinant DNA techniques. Three all with an alteration in either the vif, vpx, vpr, or nef gene were infectious to CD4-positive cell lines. The susceptibility of macaque monkey peripheral blood mononuclear cells (PBMC) infection these chimeras was examined vitro. Macaque PBMC supported replication wild-type mutant strains. A chimera carrying long terminal...

A mutant of human immunodeficiency virus type 1 which carries a frameshift insertion in the integrase/endonuclease region pol gene was constructed vitro. Upon transfection into cells, although this exhibited normal phenotype with respect to expression gag, pol, and env genes generation progeny virions, no replication-competent CD4-positive cells emerged. An assay for single-step replication defective viral genome dependent on trans complementation by rev protein established used monitor...

A highly sensitive single-round infection assay using a bacterial chloramphenicol acetyltransferase was developed to analyze an early stage of human immunodeficiency virus type 1 replication. By combination transfection and assay, with vif mutation, depending on host cells from which the derived, demonstrated be defective at phase cycle. Analysis viral proteins synthesized in indicated that incorporation Env surface protein into virions mutant, again cell-dependent way, greatly restricted....

Emerging evidence has suggested that circulating microRNAs (miRNAs) in body fluids have novel diagnostic and prognostic significance for patients with malignant diseases. The lack of useful biomarkers is a crucial problem bone soft tissue sarcomas; therefore, we investigated the miRNA signature its clinical relevance osteosarcoma.Global profiling was performed using patient serum collected from discovery cohort osteosarcoma controls cell culture media. secretion detected miRNAs cells levels...

We have discovered a novel compound, NO-1886, which possesses powerful lipoprotein lipase (LPL) activity-increasing action. Administration of NO-1886 increased LPL activity in the postheparin plasma, adipose tissue, and myocardium rats, produced reduction plasma triglyceride levels with concomitant elevation HDL cholesterol levels. enzyme mass mRNA epididymal it was concluded that mode action this compound is stimulation tissue synthesis. also conducted long-term studies to assess impact...

Lipoprotein lipase (LPL) is a key enzyme in the hydrolysis of triglyceride-rich lipoproteins. To determine role LPL lipoprotein metabolism, we established three transgenic mouse lines overexpressing human gene; highest expressor line, which transcribed mRNA heart, skeletal muscles, and adipose tissue, was used this study. The mice had 5-and 1.7-fold higher activity tissue post-heparin plasma, respectively.Plasma triglyceride levels were 24.2% that control mice, gel filtration chromatography...

The DNA sequences were determined for the lipoprotein lipase (LPL) gene from five unrelated Japanese patients with familial LPL deficiency. results demonstrated that all are homozygotes distinct point mutations dispersed throughout gene. Patient 1 has a G-to-A transition at first nucleotide of intron 2, which abolishes normal splicing. 2 nonsense mutation in exon 3 (Tyr61----Stop) and patient 8 (Trp382----Stop). latter emphasizes importance carboxy-terminal portion enzyme expression...

Apo E plays an important role in plasma lipoprotein metabolism through its high affinity binding to cell surface LDL receptor. In the present study, we studied effects of apo on atherogenic process Watanabe heritable hyperlipidemic rabbits which are deficient receptor and animal model for familial hypercholesterolemia. We isolated from 1% cholesterol-fed administered 10 mg purified intravenously into five three times a week their age 2.5 months 11 8.5 months. After sustained administration...

To ascertain the involvement of galanin in nociceptive transmission spinal dorsal horn, we examined effects intrathecal injections porcine and its antiserum on behavioral responses rat, using paw-pressure paw-radiant heat tests. An injection against reversed decreased threshold for mechanical, but not thermal, stimulation carrageenintreated hind paw with no effect non-inflamed paw. Rat as well suppressed binding [125I]porcine to antiserum, a finding suggesting that can bind rat galanin. (0.1...

We have previously reported a Trp382 (TGG) -+ stop (%A) mutation that causes familial lipoprotein lipase (LPL) deficiency.Expression study of the mutant showed truncated LPL was catalytically inactive with marked reduction in expressed mass.To investigate minimal amino-terminal sequence human required for an active enzyme, series carboxy-terminal (C-terminal) LPLs were vitro, and enzyme activity, mass, mRNA levels analyzed. The lipolytic activity stepwise between LPL-437 stop; 68% normal...

The causative mechanisms of hypertension were investigated by studying the renal function (pressure-natriuresis) curve in patients with primary aldosteronism (n = 6) and renovascular 6). Before after radical operation (removal adenoma percutaneous transluminal angioplasty hypertension), dietary NaCl intake was altered from 10 to 13 g/day Week 1 3 2. Two macaque monkeys were inoculated with a chimeric human and simian immunodeficiency virus carrying the tat, rev, vpu env genes of type 1. Infectious was recovered from one at 2 6 weeks post-infection. The hybrid nature isolated viruses verified by Southern Western blotting analyses. Both infected chimera elicited humoral antibody response against virus.

We experienced a case of reexpansion pulmonary edema (RPE) after surgical treatment pneumothorax. In this case, protein leakage and polymorphonuclear leukocyte (PMN) accumulation were observed in the reexpanded lung. Interleukin-8 leukotriene B4 fluid increased at onset RPE. PMN elastase was also increased, though its peak delayed.
