The adhesion and migration of human diploid fibroblasts on plasma clots were measured. role fibronectin was examined by depleting before clotting. Fibronectin not essential for cell spreading, although rates slightly slower depleted clots. Rates the surface unaffected depletion. In contrast, an absolute requirement cells into Cells migrated rapidly control but completely failed to penetrate fibronectin-depleted effect depletion could only be reversed adding Adsorption after clotting reverse...

Proteoglycan was isolated from cartilage and freed contaminating glycoproteins hyaluronic acid. The macromolecule consists of a protein core covalently linked to number glycosaminoglycan side chains, namely chondroitin sulphate keratan sulphate. This proteoglycan retards the attachment variety cell types tissue culture plastic collagen. Glycosaminoglycans alone, have no significant effect on rates attachment. Similarly, trypsinized is without effect. It concluded that structural integrity...

Sephacryl S-300 gel filtration of animal sera is used to demonstrate that there are at least 2 components promote the 'spreading' cells in culture. A morphometric technique described quantitate spreading process. For a number cell strains and established lines more quantitatively significant factor not fibronectin. Rather it component with fibronectin seems cause via different mechanism from stimulated by Thus will absence protein synthesis, whereas smaller requires synthesis. The kinetics...

ABSTRACT A morphometric assay has been used to show that speed of cell spreading is sensitive the level serum in surrounding growth medium. There was an inverse relationship between concentration and spreading; as increased from 0·1% 100% cells took progressively longer become fully spread. The time taken achieve complete concentrations 0·1, l·0, 10 45, 60, 105 240 min, respectively. At 3% above, fibronectin depletion had no effect on rates spreading. Only at below there although became...

Synovial fluid from rabbit elbow, shoulder, hip and knee was analysed to establish the normal levels of parameters relevant exchange ‐ viz. mass per joint, hydraulic pressure, temperature, colloid osmotic pressure (COP), protein, albumin glycosaminoglycan (GAG) concentrations. Fluid greatest in least congruous shoulder (43 ± 4 mg), cf. 6 2 mg highly hip. In (24 mg) mean thickness layer calculated be 30 µm. subatmospheric all joints (mean ‐2·8 0·4 cmH O, ‐5·7 0·3 0, knee), as many other...

A foetal-calf-serum component has been isolated that stimulates cells in culture to adopt typical fibroblast morphology. This "spreading" factor is a glycoprotein with mol. wt. of approx. 62000 and contains 12% carbohydrate; it an isoelectric point 4.8. The method described allows purification homogeneity as judged by sodium dodecyl sulphate/polyacrylamide-gel electrophoresis analytical focusing. Under cell-culture conditions the active at 18 micrograms/ml.

Human diploid fibroblasts were seeded onto or into plasma clots and different aspects of cell adhesion migration measured. The roles plasminogen activators plasmin studied by either the removal from prior to clotting addition 10 mM epsilon-aminocaproic acid, which brings about an inhibition in this system. When cells surface clots, rates attachment, spreading, unaffected depletion inhibition. In contrast, when then, although spreading unaffected, was abolished rate measured, there absolute...

The typical elongated bipolar morphology of baby hamster kidney (BHK) cells is not shown by polyoma-transformed BHK (Py-BHK) cells. Instead, the transformed line adheres poorly to tissue-culture plastic and have a more rounded than parent line.
