Abstract In this study, we evaluated the chemical chaperone activity of synthetic 4-phenylbutyric acid (4-PBA) derivatives. These derivatives have a methoxy group at benzene ring and/or longer or shorter fatty portions. Several 4-PBA demonstrated higher antiaggregation than 4-PBA. Moreover, 4-(4-methoxyphenyl)butanoic (7b) showed protective effects against endoplasmic reticulum stress-induced neuronal cell death.

Regioselectivity in Mannich reaction of 4-, 3-, and 2-substituted phenols with typical heterocyclic amines are investigated under conditions developed by us.Phenol 4-alkyl, 4-chlorophenols the title predominantly gave corresponding 2-(aminomethyl)phenols, while 4methoxyphenol afforded, addition to mono(aminomethyl)phenols, a considerable amount bis adducts.Peculiarly enough, 3-methylphenol afforded 3-methyl-4-(aminomethyl)phenols whereas 2-methylphenol produced 2-methyl-6-(aminomethyl)phenols.
