Summary. Bone marrow graft rejection following HLA‐matched bone transplantation (BMT) for leukaemia has been a rare problem. However, with the introduction of T‐lymphocyte depleted BMT, is recognized as new complication. At Royal Free Hospital (RFH) in London T‐depletion achieved using two monoclonal antibodies complement mediated lysis. The methodology was extended to other centres and total 56 patients have received T‐depleted, HLA matched BMT. Twelve had rejection. RFH three 41 (7%)...

Mortality resulting from influenza (flu) virus infections occurs primarily in the elderly through declining immunity. Studies mice have suggested beneficial effects of selenium (Se) supplementation on immunity to flu but similar evidence is lacking humans. A dietary intervention study was therefore designed test Se-supplementation a variety parameters anti-flu healthy subjects aged 50-64 years.A 12-week randomized, double-blinded, placebo-controlled clinical trial...

Changes in the composition of gut microbiota have been implicated pathogenesis allergic disorders, suggesting beneficial interactions between intestinal immune system and specific bacterial strains. Lactobacilli are naturally present within complex gastrointestinal humans they currently many probiotic supplements. We sought to investigate role that Lactobacillus casei Shirota (LcS) may play modulating seasonal rhinitis (SAR).The study format was double-blinded, placebo-controlled with 10 SAR...

Abstract Using three-colour flow cytometry, we have measured intracellular IL-2, interferon-gamma (IFN-γ) and tumour necrosis factor-alpha (TNF-α) induced in human CD4+ CD8+ T cells from normal donors patients with common variable immunodeficiency (CVID). Since a new range of directly FITC-conjugated anti-cytokine antibodies was used, conditions were optimized for the concentration antibody, cell permeabilization fixation, time exposure to monensin retain cytokines within cell. Kinetics...

Objective — We hypothesized that acute hyperglycemia (an independent cardiovascular risk factor) increases the expression of proatherogenic leukocyte adhesion molecule in type 2 diabetes and controls these molecules would be antioxidant sensitive. Methods Results Twenty-three patients 13 control underwent two oral glucose tolerance tests 14 days apart took placebo or 800 IU daily alpha tocopherol between tests. Monocyte neutrophil Mac-1, LFA-1 3, ICAM-1, VLA-4 were measured at 0, 120, 240...

Abstract We have compared CD10 antigen expression in normal fetal bone marrow with that of B‐lineage acute lymphoblastic leukemia (ALL). Both quantitative indirect immunofluoresence (QIFI) and direct immunofluorescence (IF) tests Quantum beads were used to convert median fluorescence intensity (MFI) values into numbers molecules expressed per cell (AgE). Lymphoid precursors the liver 3–12.5 × 10 3 molecules/cell an upper limit 5 4 /cell (MaxAgE). The AgE different cases ALL variable ranged...

Abstract Epidemiological studies have shown that the risk of developing oesophageal adenocarcinoma (OA) is inversely correlated to consumption fruits and vegetables. Flavan‐3‐ols are most abundant subclass flavonoids in these types foods. Three apple‐derived procyanidin fractions with different average degrees polymerization (aDP) were characterized effects pure flavan‐3‐ol monomers ((–)‐epicatechin (+)‐catechin) dimers (B1, B2) on two OA cell lines investigated. Flavan‐3‐ol had no effect...

Objective To determine effects of probiotic consumption on clinical and immunological parameters seasonal allergic rhinitis (SAR) in an out-of-season single nasal allergen challenge. Methods In a study registered at ClinicalTrials.Gov (NCT01123252), 16-week dietary intervention was undertaken 60 patients with (>16 years old). Using double-blinded, placebo-controlled anonymised design, the were divided equally into two groups. One group given dairy drink containing Lactobacillus casei Shirota...

Abstract Alloantigens, unlike recall antigens, activate both CD45RA + (naive) and CD45RO (memory) CD4 cells to the same extent. These T cell subsets may therefore interact with each other in response alloantigens on transplanted grafts. We have investigated if ability of activated produce respond interleukin 2 (IL2) IL4 be involved this interaction. After activation, up‐regulate their IL2 receptor (IL2R) IL4R expression, yet substantially enhanced proliferation but not subset, while...

Introduction: Ageing is accompanied by increased susceptibility to infection and age-associated chronic diseases. It also associated with reduced vaccine responses, which often attributed immunosenescence the functional decline of immune system. Immunosenescence characterized a chronic, low-grade, inflammatory state termed inflammaging. Habitants Mediterranean regions maintain good health into old age; diets. Hypothesis: Adoption (MED)-diet elderly subjects, in Norfolk (UK), may improve...
