Two-cysteine peroxiredoxins (Prx) have a three-step catalytic cycle consisting of (1) reduction peroxide and formation sulfenic acid on the enzyme, (2) condensation with thiol to form disulfide, also known as resolution, (3) disulfide by reductant protein. By following changes in protein fluorescence, we studied pH dependence reaction 2 human 1, 2, 5 Salmonella typhimurium AhpC obtained rate constants for pKa values involved each system. The observed constant spans orders magnitude, but all...

Abstract Peroxiredoxins are thiol‐dependent peroxidases that function in peroxide detoxification and H 2 O induced signaling. Among the six isoforms expressed humans, PRDX1 PRDX2 share 97% sequence similarity, 77% identity including active site, subcellular localization (cytosolic) but they hold different biological functions albeit associated with their peroxidase activity. Using recombinant human PRDX2, kinetics of oxidation hyperoxidation peroxynitrite were followed by intrinsic...

Stevia species (Asteraceae) have been a rich source of terpenoid compounds, mainly sesquiterpene lactones, several which show antiprotozoal activity. In the search for new trypanocidal S. satureiifolia var. and alpina were studied. Two santhemoidin C 2-oxo-8-deoxyligustrin, respectively, isolated. These compounds assessed in vitro against Trypanosoma cruzi stages, showing IC50 values 11.80 4.98 on epimastigotes, 56.08 26.19 trypomastigotes 4.88 20.20 µM amastigotes, respectively....

Abstract This practical class activity was designed to introduce students recombinant protein expression and purification. The principal goal is shed light on basic aspects concerning production, in particular expression, chromatography methods for purification, enzyme as a tool evaluate purity conformation of the product. Herein, we describe purification glutathione transferase from human parasite Echinococcus granulosus (EgGST1), causative agent hydatidosis. EgGST1 expressed fused...
