The non-proteinogenic tris-amino acid labionin was discovered in 2010 and since has been described many natural products, defining an entire family of lanthipeptides. unusual amino is biosynthetically produced by the sequential condensation a cysteine onto two dehydroalanines. An attempt 2011 to synthesize monomeric unit unsuccessful yet served as valuable foundation guide present work. A recently disclosed methodology for selective opening aziridines mercaptan nucleophiles applied another...

Abstract Increasing the speed, specificity, sensitivity, and accessibility of mycobacteria detection tools are important challenges for tuberculosis (TB) research diagnosis. In this regard, previously reported fluorogenic trehalose analogues have shown potential, but their green‐emitting dyes may limit sensitivity applications in complex settings. Here, we describe a trehalose‐based probe featuring molecular rotor turn‐on fluorophore with bright far‐red emission (RMR‐Tre). RMR‐Tre, which...

Lantibiotics are a class of peptide antibiotics with activity against most Gram-positive bacteria. Lanthionine (Lan) and β-MeLan unusual thioether-bridged, non-proteinogenic amino acids, which characteristic features lantibiotics. In this paper, we report the facile stereoselective synthesis β-methyllanthionines orthogonal protection by nucleophilic ring opening aziridines. This method leads to an expedient access allows production over 30 g β-methyllanthionine in single batch.

ABSTRACT Macrodomains are a class of conserved ADP-ribosylhydrolases expressed by viruses pandemic concern, including coronaviruses and alphaviruses. Viral macrodomains critical for replication virus-induced pathogenesis; therefore, these enzymes promising target antiviral therapy. However, no potent or selective viral macrodomain inhibitors currently exist, in part due to the lack high-throughput assay this enzymes. Here, we developed ADP-ribosylhydrolase using SARS-CoV-2 Mac1. We performed...

Virus-like particles (VLPs) have untapped potential for packaging and delivery of macromolecular cargo. To be a broadly useful platform, there needs to strategy attaching macromolecules the inside or outside VLP with minimal modification platform Here, we repurpose antiviral compounds that bind hepatitis B virus (HBV) capsids create chemical tag noncovalently attach cargo VLP. Our consists capsid assembly modulator, HAP13, connected linker terminating in maleimide. is green fluorescent...
