Data mining of the Corynebacterium glutamicum genome identified 4 genes analogous to mshA, mshB, mshC, and mshD that are involved in biosynthesis mycothiol Mycobacterium tuberculosis smegmatis. Individual deletion these was carried out this study. Mutants mshC- mshD- lost ability produce mycothiol, but mutant mshB- produced as wild type did. The phenotypes mutants were same when grown LB or BHIS media, not able grow mineral medium with gentisate 3-hydroxybenzoate carbon sources. C....
Regulation of sulphur metabolism in Corynebacterium glutamicum ATCC 13032 has been studied intensively the last few years, due to its industrial as well scientific importance. Previously, gene cg0156 was shown belong regulon McbR, a global transcriptional repressor C. glutamicum. This encodes putative ROK-type regulator, paralogue activator sulphonate utilisation, SsuR. Therefore, it is an interesting candidate for study further understanding regulation glutamicum.Deletion cg0156, now...
Mycothiol (MSH) functions as major low molecular weight (LMW) thiol in the industrially important Corynebacterium glutamicum. In this study, we genomically integrated an Mrx1-roGFP2 biosensor C. glutamicum to measure dynamic changes of MSH redox potential (EMSH) during growth and under oxidative stress. maintains a highly reducing intrabacterial EMSH throughout curve with basal levels ~− 296 mV. Consistent its H2O2 resistant phenotype, responds only weakly 40 mM H2O2, but is rapidly oxidized...
Three novel corynebacterial species were isolated from soil sampled at a paddock in Vilsendorf, North Rhine-Westphalia, Germany. The strains coccoid or irregular rod-shaped, catalase-positive and pale white to yellow-orange colour. By whole genome sequencing comparison of the 16S rRNA genes as well structure, it was shown that all three represent family Corynebacteriaceae, order Corynebacteriales, class Actinobacteria. This project describes isolation, identification, sequencing, phenotypic...
