ABSTRACT Diosmin (DSN) is found mainly in citrus fruits, and has potent antioxidant effects. This study aimed to evaluate pharmacokinetics of diosmetin-7-glucoside-γ-cyclodextrin (DIOSG-CD) inclusion complex. The area under the curve values from AUC0-24 DIOSG-CD, prepared by reacting DSN naringinase with γ-CD, were approximately 800-fold higher than those following their administration Sprague-Dawley rats.

Ginkgo biloba extract (GBE) is an essential herbal supplement for diverse health perspectives. The present study aims to improve the oral bioavailability of GBE flavonoids. We formulated flavonoids-cyclodextrin (GBE-CD) complex by reacting flavonoids with naringinase in presence γ-cyclodextrin (CD) using a patented method. results showed enhanced quercetin (QCT), kaempferol (KAE), and isorhamnetin (ISR) after β-glucuronidase catalysis flavonoid glucuronide conjugates plasma GBE-CD (25...

Flavonoids such as quercetin and its glucosides, especially isoquercitrin are well known anti-inflammatory, anti-allergic, anti-carcinogenic, etc. The safety of formulations needs to be established prior their use in functional food applications. mutagenicity genotoxicity the IQC-γCD inclusion complex were assessed with three standard assays bacterial reverse mutation assay (Ames test) using a combined in-vivo micronucleus comet under Organisation for Economic Co-operation Development (OECD)...

Currently, revisions to the ICH S1 guidance on rodent carcinogenicity testing are being proposed. Application of this approach would reduce use animals in accordance with 3Rs principles (reduce/refine/replace). The method also shift resources focus more scientific mechanism-based assessments and promote safe ethical development new small molecule pharmaceuticals. In revised draft, findings such as cellular hypertrophy, diffuse and/or focal hyperplasia, persistent tissue injury chronic...

Abstract Background The repeated-dose liver micronucleus (RDLMN) assay has been well-developed and applied because of its simplicity the ease integration into general toxicity studies which is preferred method from 3R’s point view. In this assay, we observed micronucleated hepatocytes accumulated during a rather long-term dosing period. When considering studies, effects age animals used in becomes major issue. effect on induction rate reported bone marrow assays, it considered that decrease...

CooA, a homodimeric haem-containing protein, is responsible for transcriptional regulation in response to carbon monoxide (CO). It has b-type haem as CO sensor. Upon binding the haem, CooA binds promoter DNA and activates expression of genes metabolism. from Carboxydothermus hydrogenoformans been overexpressed Escherichia coli, purified crystallized by vapour-diffusion method. Abstract Background Di(2-ethylhexyl) phthalate (DEHP) is a plasticizer commonly used in wide variety of products, including medical devices. It rapidly metabolized the liver into various metabolites upon absorption through oral ingestion, dermal absorption, and inhalation. DEHP classified as non-genotoxic hepatocarcinogen rodents, its chronic exposure has been associated with development cancer these animals, but most genotoxicity studies have negative. Epidemiologic humans suggest that...

The carcinogenicity of 2,2'-[1,2-ethanediylbis(oxymethylene)]bis-oxirane (ethylene glycol diglycidyl ether; EGDE), 3-hydroxy-2-naphthoic acid (HNA), and acetoacetanilide (AAA) was investigated using a medium-term rat liver bioassay for an occupational safety assessment. F344 male rats were administered single intraperitoneal injection diethylnitrosamine (200 mg/kg body weight (bw)/day) then starting 2 weeks later, they received EGDE at 6, 20, 60 bw/day, HNA 60, 200 or AAA 200, 600 bw/day by...
