The intrinsically disordered regions of eukaryotic proteomes are enriched in short linear motifs (SLiMs), which crucial relevance for cellular signaling and protein regulation; many mediate interactions by providing binding sites peptide-binding domains. vast majority SLiMs remain to be discovered highlighting the need experimental methods their large-scale identification. We present a novel proteomic peptide phage display (ProP-PD) library that displays peptides representing human proteome,...

Glucokinase (GK) is a monomeric allosteric enzyme and plays pivotal role in blood glucose homeostasis. GK regulated by regulatory protein (GKRP), indirectly effectors of GKRP. Despite the critical roles GKRP, molecular basis for regulation mechanism GKRP remains unclear. We determined crystal structure Xenopus complex presence fructose-6-phosphate at 2.9 Å. binds to super-open conformation mainly through hydrophobic interaction, inhibiting activity locking small domain GK. demonstrate...

Analysis of protein dynamics using single-molecule fluorescence resonance energy transfer (smFRET) is widely used to understand the structure and function proteins. Nonetheless, site-specific labeling proteins with a pair donor acceptor dyes still remains challenge. Here we present general facile method for dual by incorporating two different, readily available, unnatural amino acids (p-acetylphenylalanine alkynyllysine) smFRET. We newly evolved alkynyllysine-specific aminoacyl-tRNA...

Cholangiocarcinoma (CCA) is a silent tumor with high mortality rate due to the difficulty of early diagnosis and prediction recurrence even after timely surgery. Serologic cancer biomarkers have been used in clinical practice, but their low specificity sensitivity problematic. In this study, we aimed identify CCA-specific glycan epitopes that can be for elucidate mechanisms by which glycosylation altered progression. The serum patients various cancers was fractioned into membrane-bound...

Studies on glutamine (Gln) metabolism have highlighted the vital role of Gln in cellular functions and its potential as a biomarker for disease detection. Despite increasing interest metabolism, in-depth evaluations are challenging owing to limitations conventional Gln-measuring methods. Thus, we developed ligand-induced dimerization-based sensor Gln, termed Q-SHINE, by splitting glutamine-binding protein into two separate domains. Q-SHINE enables highly accurate convenient measurement...

Cyanobacteriochromes (CBCRs) are linear tetrapyrrole bilin-binding photoreceptors of cyanobacteria that exhibit high spectral diversity, gaining attention in optogenetics and bioimaging applications. Several engineering studies on CBCRs were attempted, especially for designing near-infrared (NIR) fluorescent proteins with longer fluorescence wavelengths. However, despite continuous efforts, a key component regulating emission property is still poorly understood. As model system, we focused...
