Sigma factors are transcriptional regulators that part of complex regulatory networks for major cellular processes, as well growth phase-dependent regulation and stress response. Actinoplanes sp. SE50/110 is the natural producer acarbose, an α-glucosidase inhibitor used in diabetes type 2 treatment. Acarbose biosynthesis dependent on growth, making sigma factor engineering a promising tool metabolic engineering. ACSP50_0507 homolog developmental osmotic-stress-regulating Streptomyces...
Three novel corynebacterial species were isolated from soil sampled at a paddock in Vilsendorf, North Rhine-Westphalia, Germany. The strains coccoid or irregular rod-shaped, catalase-positive and pale white to yellow-orange colour. By whole genome sequencing comparison of the 16S rRNA genes as well structure, it was shown that all three represent family Corynebacteriaceae, order Corynebacteriales, class Actinobacteria. This project describes isolation, identification, sequencing, phenotypic...
As the natural producer of acarbose, Actinoplanes sp. SE50/110 has high industrial relevance. Like most Actinobacteria, strain carries several more putative biosynthetic gene clusters (BGCs) to produce further products, which are be discovered. Applying a metabolomics-guided approach, we tentatively identified five compounds that produced by strain: watasemycin, thiazostatin, isopyochelin, pulicatin, and aerugine. A comparison genomic context allowed identification BGC, is highly similar...
