Hi-res view of human Aβ42 filaments Alzheimer’s disease is characterized by a loss memory and other cognitive functions the filamentous assembly Aβ tau in brain. The peptides into that end at residue 42 central event. Yang et al . used electron cryo–electron microscopy to determine structures from brain (see Perspective Willem Fändrich). They identified two types related S-shaped filaments, each consisting identical protofilaments. These will inform development better vitro animal models,...
Abstract Many age-dependent neurodegenerative diseases, such as Alzheimer’s and Parkinson’s, are characterized by abundant inclusions of amyloid filaments. Filamentous the proteins tau, amyloid-β, α-synuclein transactive response DNA-binding protein (TARDBP; also known TDP-43) most common 1,2 . Here we used structure determination cryogenic electron microscopy to show that residues 120–254 lysosomal type II transmembrane 106B (TMEM106B) form filaments in human brains. We determined...
Soluble oligomers of amyloid β-protein (Aβ) have been defined as aggregates in supernatants following ultracentrifugation aqueous extracts from Alzheimer's disease (AD) brains and are believed to be upstream initiators synaptic dysfunction, but little is known about their structures. We now report the unexpected presence Aβ fibrils synaptotoxic high-speed AD extracted by soaking an buffer. The did not appear form during preparation, counts EM correlated with ELISA quantification. Cryo-EM...
Abstract The hydroxycarboxylic acid receptor 2 (HCA2) agonist niacin has been used as treatment for dyslipidemia several decades albeit with skin flushing a common side-effect in treated individuals. Extensive efforts have made to identify HCA2 targeting lipid lowering agents fewer adverse effects, despite little being known about the molecular basis of mediated signalling. Here, we report cryo-electron microscopy structure HCA2-G i signalling complex potent MK-6892, along crystal structures...
We successfully achieved the phosphorylation of secondary aromatic alcohols with H-phosphine oxides (less developed system) using phosphotungstic acid as a catalyst in dimethyl carbonate. The system was simple and environmentally friendly showed better activity than traditional Lewis or Brønsted acids such FeCl3, p-TsOH·H2O, etc., generating up to 97% isolated yield. Control experiments indicated that reaction did not occur through radical pathway, ethers carbocation were key intermediates pathway.
<title>Abstract</title> Gene dosage and point mutations in SNCA, the a-synuclein gene, give rise to familial forms of Parkinson’s disease dementia with Lewy bodies; an insertion mutation SNCA causes juvenile-onset synucleinopathy. We previously reported electron cryo-microscopy (cryo-EM) structures filaments from brains individuals disease, bodies multiple system atrophy, as well brain individual Here we report cryo-EM frontal cortex two cases Parkinsonism...
Endometriosis, characterized by extrauterine endometrial tissue, leads to irregular bleeding and pelvic pain. Menstrual retrograde theory suggests fragments traverse fallopian tubes, causing inflammation scar tissue. Prevalent among infertile women, risk factors include fewer pregnancies, delayed childbirth, cycles, familial predisposition. Treatments, medication, surgery entail side effects. Studies link gut microbiota alterations endometriosis, necessitating research establish causation....
Abstract Alzheimer’s Disease (AD) is the most common cause of dementia afflicting 55 million individuals worldwide, with limited treatment available. Current AD models mainly focus on familial (fAD), which due to genetic mutations. However, for studying sporadic (sAD), represents over 95% cases without specific mutations, are severely limited. Moreover, fundamental species differences between humans and animals might significantly contribute clinical failures therapeutics that have shown...
Abstract Soluble aggregates of amyloid-β (Aβ), often called oligomers, are believed to be principal drivers neurotoxicity, spreading pathology, and symptoms in Alzheimer’s disease (AD), but little is known about their structures human brain. Aβ oligomers have been defined as found supernatants following ultracentrifugation aqueous extracts. We now report the unexpected presence abundant fibrils high-speed from AD brains that were extracted by soaking buffer. The did not appear form during...
To investigate the correlation between serum levels of 25-hydroxy vitamin D [25(OH)D] and visceral fat area patients with type 2 diabetes mellitus (T2DM) in context different bone mass.A total 180 T2DM were randomly selected for mineral density (BMD) examination. According to results, they divided into three groups: normal group (group A); mass reduction B); osteoporosis C).Serum 25(OH)D NC group, A B C decreased turn, Visceral (VFA) significantly higher than those [(29.41±4.87) vs....
Phosphotungstic acid with a Keggin structure as an efficient, simple and green catalyst for the phosphinylation of secondary propargyl alcohols phosphine oxides to afford γ-ketophosphine up 88% isolated yield was developed using dimethyl carbonate solvent. Diaryl- or alkylaryl-substituted alcohols, diaryl arylalkylphosphine could tolerate system, which reduced dosage, avoided use multi-components toxic solvents. More interestingly, phosphotungstic exhibited best activity when 0.58 moles...
Objective: To explore the clinical value of von Willebrand Factor (vWF) and VITRO score (vWF:Ag/platelet count) in assessing disease progression patients with HBV infection. Methods: Randomly collect relevant data 308 infection (including 154 cases chronic hepatitis B, 66 B cirrhosis compensatory period, 88 decompensated period) from December 1, 2018 to January 5, 2021 Second Affiliated Hospital Chongqing Medical University. The vWF values are measured by a uniform optical method, all...
To compare the therapeutic effects between sitagliptin and voglibose both with sensor-augmented insulin pump (SAP) in newly diagnosed type 2 diabetes mellitus (T2DM).Fifty-six hospitalized T2DM patients Department of Endocrinology First Affiliated Hospital Dalian Medical University, hemoglobin A1c (HbA1c) value 9%-11%, were randomized into (S) group (n=28) (V) by block randomisation. Participants S received 100 mg per day, V 0.2 for 3 times day. All treated SAP 9 days. Real-time continuous...
