Purposes Lymphopenia is extensively studied, but not circulating leucocyte subpopulations, which however have distinct roles in tumor tolerance. Proton therapy has been shown to a lesser impact on the immune system than conventional X-ray radiotherapy through lower dose exposure healthy tissues. We explored differential effects of brain and proton irradiation subpopulations.
VDJ recombination of T cell receptor and immunoglobulin loci occurs in immature lymphoid cells. Although the molecular mechanisms DNA cleavage ligation have become more clear, it is not understood what controls which target undergo rearrangement. In interleukin 7 (IL-7R)alpha-/- murine thymocytes, has been shown that rearrangement (TCR)-gamma locus virtually abrogated, whereas other rearranging are less severely affected. By examining different strains mice with targeted mutations, we now...
To ascertain how the actual repertoire of T cell receptors (TCRs) deviates from theoretical, we have generated a large number junctional region sequences TCRs carrying V beta 17 variable region. The greater than 600 analyzed represent transcripts nine different populations, permitting several comparisons: an expressed vs. non-expressed allele, those E+ E- mice, immature mature thymocytes, thymic peripheral cells, and CD4+ CD8+ cells. These comparisons allowed us to distinguish between...
Nonobese diabetic (NOD) mice spontaneously develop a T-cell-mediated autoimmune disease that is similar in many respects to insulin-dependent diabetes mellitus humans. T-cell clones specifically recognize pancreatic islet cell antigens can be derived from NOD mice, and most of these have been diabetogenic upon transfer healthy recipients. We report herein the sequences receptor alpha beta chains four NOD-derived, islet-specific clones. The are quite heterogeneous--in junctional regions,...
Though radiotherapy is a local therapy, it has systemic effects mainly influencing immune and inflammation processes. This important consequences in the long-term prognosis therapy individualization. Our objective was to investigate inflammation-related changes peripheral blood of head neck cancer patients treated with radiotherapy. Peripheral cells, plasma cell-derived RNA were isolated from 23 before at two time points after cellular parameters, protein gene expression alterations studied....
Although lymphopenia is linked to immune suppression favoring tumor growth, the effect of different radiation types on specific T-lymphocyte subsets remains unclear. Among subpopulations, CD8+-lymphocytes serve as key effectors in cancer immunity. This study aimed examine changes subpopulations both tumor-free and glioblastoma-bearing mice following brain-irradiation. The was divided into two main phases. First, C57BL/6 mice, with or without glioblastoma (GL261 cells), received hemispheric...
RJ 2.2.5 is an immunoselected mutant of the Burkitt lymphoma line Raji. It fails to display MHC class II antigens at cell surface due a transcriptional defect. We have identified function regulatory factor that defective in cells; this absolutely required for activity gene enhancer.
Foxp3 + regulatory T cells (Tregs) in the colon are key to promoting peaceful coexistence with symbiotic microbes. Differentiated either thymic or peripheral locations, and modulated by microbes other cellular influencers, colonic Treg subsets have been identified through transcription factors (TFs; Helios, Rorγ, Gata3, cMaf), but their interrelationships unclear. Applying a multimodal array of immunologic, genomic, microbiological assays, we find more overlap than expected between...
Mutagenic and carcinogenic UV-B radiation is known to damage DNA mostly through the formation of bipyrimidine photoproducts, including cyclobutane dimers (CPD) (6-4) photoproducts ((6-4) PP). Using high-performance liquid chromatography coupled tandem mass spectrometry, we investigated repair thymine-thymine (TT) thymine-cytosine (TC) CPD PP in cultured human dermal fibroblasts. A major observation was that rate did not depend on identity modified pyrimidines. In addition, removal found...
Abstract IL-7, a cytokine produced by thymic epithelium, was shown to induce adhesion of murine thymocytes gelatin-coated membranes. A major binding component gelatin identified as fibronectin. IL-7-induced observed for all the thymocyte subsets, including double-negative, double-positive, and single-positive cells, specific IL-7R were verified on each subset. Fibronectin mediated via alpha4beta1 integrin (VLA-4), which is expressed at high levels thymocytes. VLA-4 surface expression not...
