Multipotent adult resident cardiac stem cells (CSCs) were first identified by the expression of c-kit, cell factor receptor. However, in myocardium c-kit alone cannot distinguish CSCs from other c-kit-expressing (c-kitpos) cells. The heart indeed contains a heterogeneous mixture c-kitpos cells, mainly composed mast and endothelial/progenitor This heterogeneity has generated confusion controversy about existence role heart. Here, to unravel CSC identity within population, separated through...
Vascularization is driven by morphogen signals and mechanical cues that coordinately regulate cellular force generation, migration, shape change to sculpt the developing vascular network. However, it remains unclear whether vasculature actively regulates its own properties achieve effective vascularization. We engineered tissue constructs containing endothelial cells fibroblasts investigate mechanics of Tissue stiffness increases during morphogenesis resulting from emergent interactions...
Abstract A key event in the formation of pharyngeal arches is outpocketing endodermal pouches and establishment contact with overlying ectoderm. However, relatively little known about how endoderm ectoderm relate to each other at these points extent which this differs between pouches. We have therefore detailed interactions chick embryo. Unlike pouches, first pouch does not sustain direct but separates after initial contact. Contrastingly, a perforation formed second cleft that creates an...
The development of cellular therapies to treat muscle wastage with disease or age is paramount. Resident satellite cells are not currently regarded as a viable cell source due their limited migration and growth capability ex vivo. This study investigated the potential muscle-derived PW1+/Pax7– interstitial progenitor (PICs) tissue-specific stem/progenitor stem properties multipotency. Sca-1+/PW1+ PICs were identified on tissue sections from hind limb 21-day-old mice, isolated by...
Pharyngeal segmentation is a defining feature of vertebrate embryos and apparent as series bulges found on the lateral surface embryonic head, pharyngeal arches. The ancestral condition for gnathostomes to have seven segments: jaw, hyoid, five posterior branchial However, within sarcopterygians, region has undergone extensive remodelling that resulted in reduction number segments, such amniotes only aim this study probe developmental basis loss segments. We therefore compared development...
Skeletal muscle-derived PW1pos/Pax7neg interstitial cells (PICs) express and secrete a multitude of proregenerative growth factors cytokines. Utilizing porcine preclinical skeletal muscle injury model, delivery allogeneic PICs (pPICs) significantly improved accelerated myofiber regeneration neocapillarization, compared with saline vehicle control-treated muscles. Allogeneic pPICs did not contribute to new myofibers or capillaries were eliminated by the host immune system. In conclusion, pPIC...
