Direct force measurements by atomic microscopy (AFM) have become an indispensable analytical tool in the last decades. Force been widely used for adhesion measurements, often combination with colloidal probe technique. For latter technique, a particle is attached to end of AFM cantilever, proving great flexibility terms colloid/surface interaction be studied. Interestingly, question how much data necessary obtain statistically reliable results has addressed this context only sparsely. By...
In the early process development, feeding trajectories and cultivation conditions are altered in order to maximise desired product. This is contrast industrial cultivations, where variations be eliminated. fully understand safely run a new process, real-time information beneficial. Near-infrared spectrometer that coupled fermenter offer possibility gather on-line. The NIR data can transformed via partial least squares modeling estimate substrate biomass concentrations. At low concentrations...
In this study, we show the successful application of different model-based approaches for maximizing macrolactin D production by Paenibacillus polymyxa. After four initial cultivations, a family nonlinear dynamic biological models was determined automatically and ranked their respective Akaike Information Criterion (AIC). The best were then used in multi-model setup robust product maximization. experimental validation shows highest yield attained compared with identification runs so far....
This paper presents a case study on integrating new model extension describing the production of enzyme soluble hydrogenase (SH) into an already existing by means on-line optimal experimental design (OED). These investigations were done autotrophic cultivation system and process Ralstonia eutropha, i.e., where aeration air is replaced gas consisting H2/O2/CO2. Prior to experiment, three different structures are postulated coarsely identified. Off-line OED with best yields initial feeding...
Abstract Although known to be very powerful, the widespread application of model‐based techniques is still significantly hampered in area bio‐processes. Reasons for this situation can found along whole chain set up and implement such approaches. In a time‐consuming step, models are typically hand‐crafted. Whether alternatives better exist actually fulfill final goals undocumented, most often even unknown. next process control methods hand‐coded an error‐prone procedure. For many these given...
