This work presents one-incubation one-hour multiplex ELISA enabled by aqueous two-phase systems for five-plex cytokine detection in human ThP-1 macrophages.
Microplate titration quantifies sodium hydroxide generated from formaldehyde reacting with excess sulfite in a 96-microwell plate. Phenolphthalein indicators change red to colorless when all ions react. Methodology optimized reagent concentrations, and reaction time created Calibration Chart for semi-quantitative determination.
