We report biodegradable plasmon resonant liposome gold nanoparticles (LiposAu NPs) capable of killing cancer cells through photothermal therapy. The pharmacokinetic study LiposAu NPs performed in a small animal model indicates situ degradation hepatocytes and further getting cleared the hepato-biliary renal route. Further, therapeutic potential tested mouse tumor xenograft using NIR laser (750 nm) illumination resulted complete ablation mass, thus prolonging disease-free survival.
Infection of the genitourinary tract with Group B Streptococcus (GBS), an opportunistic gram positive pathogen, is associated premature rupture amniotic membrane and preterm birth. In this work, we demonstrate that GBS produces vesicles (MVs) in a serotype independent manner. These MVs are loaded virulence factors including extracellular matrix degrading proteases pore forming toxins. Mice chorio-decidual membranes challenged ex vivo resulted extensive collagen degradation leading to loss...
The induction of durable protective immune responses is the main goal prophylactic vaccines, and adjuvants play a role as drivers such responses. Despite advances in vaccine strategies, safe effective HIV remains significant challenge. use an appropriate adjuvant crucial to success vaccines. Here we assessed saponin/MPLA nanoparticle (SMNP) with envelope (Env) trimer, evaluating safety impact multiple variables including dose (16-fold range), immunization route, composition on establishment...
Type I interferons (IFN-I) are critical mediators of innate control viral infections but also drive the recruitment inflammatory cells to sites infection, a key feature severe coronavirus disease 2019. Here, IFN-I signaling was modulated in rhesus macaques (RMs) before and during acute SARS-CoV-2 (severe respiratory syndrome 2) infection using mutated IFN-α2 (IFN-modulator; IFNmod), which has previously been shown reduce binding endogenous IFN-I. IFNmod treatment uninfected RMs observed...
Dual stimuli pH and temperature-responsive nanohydrogels based on poly(N-isopropylacrylamide)-chitosan have been synthesized. Fe3O4 magnetic nanoparticles (NPs) (-12 nm) incorporated into hydrogels to achieve temperature optimized nanohydrogel (MNHG) for hyperthermia with lower critical solution temperature, LCST > 42 degrees C. The composite was further investigated its potential application in drug delivery vitro cancer cell cytotoxicity. Water-bath assisted release studies were carried...
The pathological aggregation of tau is one the major contributing factors for several neurodegenerative tauopathies, including Alzheimer's disease. Here, we report that C1, a synthetic derivative curcumin, strongly inhibited both and filament formation purified protected neuroblastoma cells from deleterious effects oligomers. Using confocal microscopy, C1 was found to reduce size number droplets increased critical concentration required droplet in vitro indicating suppressed liquid-liquid...
This study reports a hybrid lipo-polymeric nanosystem (PDPC NPs) synthesized by modified hydrogel-isolation technique. The ability of the to encapsulate hydrophilic and hydrophobic molecules has been demonstrated, their enhanced cellular uptake observed in vitro. PDPC NPs, surface coated with gold situ reduction chloroauric acid (PDPC-Au NPs), showed photothermal transduction efficacy ∼65%. PDPC-Au NPs demonstrated an increase intracellular ROS, triggered DNA damage resulted apoptotic cell...
A protective vaccine against HIV will likely need to induce broadly neutralizing antibodies (bnAbs) that engage relatively conserved epitopes on the envelope glycoprotein (Env) trimer. Nearly all strategies bnAbs require use of complex immunization regimens involving a series different immunogens, most which are Env trimers. Producing protein-based clinical material evaluate such in humans presents major challenges cost and time. Furthermore, with trimers as soluble proteins induces strong...
Abstract Bioactive nanosilicates are emerging prominent next generation biomaterials due to their intrinsic functional properties such as advanced biochemical and biophysical cues. Recent studies show interesting dose-dependent effect of fluoride ions on the stem cells. Despite well nanosilicate, there is no reported literature fluoride-doped We have systematically evaluated interaction nanosilicate platelets (NS + F) with human dental follicle cells (hDFSCs) probe cytotoxicity, cellular...
Cancer stem-like cells (CSCs) have emerged as an important target for breast cancer therapy owing to their self-renewability, proliferation, and elevated chemoresistance properties. Here, we present a strategy of eliminating CSCs by differentiation where "forced differentiation" reprograms so that they lose intrinsic properties become susceptible conventional chemotherapeutic drugs. In this study, report paclitaxel enhances the stemness CSCs, while phytochemical forskolin being essentially...

Photothermal therapy (PTT) is emerging as an effective treatment modality for cancer due to its noninvasive nature. However, the pro-inflammatory necrotic cell death during PTT limits successful clinical application. Here, we have developed quercetin (QE)-loaded biodegradable plasmonic nanoparticles that can specifically induce apoptosis in cells after PTT. We synthesized gold-coated liposome (LiposAu) and QE-loaded (QE-LiposAu) by situ reduction of chloroauric acid with ascorbic presence...
Therapeutic protein depots have limited clinical success because of the presence critical preparation barriers such as low encapsulation, uncontrolled release, and activity loss during processing storage. In present study, we used our novel protein-nanoencapsulation (into sugar-glass nanoparticle; SGnP) platform to prepare a depot overcome abovementioned formidable challenges. The SGnP-mediated microparticle has been validated using four model proteins (bovine serum albumin, horseradish...
Shape-directing agents that promote anisotropic growth are frequently employed in the synthesis of gold nanorods (GNRs), a typical example which is surfactant cetyltrimethylammonium bromide (CTAB). Owing to their cytotoxicity, surfactant-passivated GNRs have little use biological applications unless made biocompatible via additional downstream processing. Reported herein first instance liposome-directed synthesized absence surfactants. The as-synthesized phospholipid-passivated readily...
Photothermal therapy (PTT) is one of the emerging modalities for treatment cancer. Among various near Infrared (NIR) responsive nanomaterials which are under research, gold coated liposomes (Au Lipos NPs) considered as a promising candidate photothermal and triggered drug delivery agent. The external NIR light trigger advantageous because itself causes therapeutic effect (by inducing localized hyperthermia). In current study we have investigated timing that governs efficacy. We found...
SUMMARY Rare B cells can have special pathogen-recognition features giving them the potential to make outsized contributions protective immunity. However, rare naive infrequently participate in immune responses. We investigated how germline-targeting vaccine antigen delivery and adjuvant selection affect priming of exceptionally BG18-like HIV broadly neutralizing antibody-precursor (~1 50 million) non-human primates. Only escalating dose (ED) immunization using saponin SMNP elicited...
